Objective:1. To establish a method for the isolation and culture of SD rat adipose-derivedstem cells (ADAS) in vitro, and explore their proliferation ability and biological characteristics. Also to test and verify the cell surface markers of ADSCs.2. To demonstrate the multilineage differentiation potentials of ADSCs and explore the capability of ADSCs differentiate into osteoblasts. To valuate the feasibility of using ADSC as a new cell source for bone engineering.3. To amplificate and purify what we have constructed replication-incompetent adenovirus vectors carrying hBMP-2(human bone morphogenetic protein-2) gene and GPF(green fluorescent protein) gene respectively , and to mensurate the vector.To study the effects of transfection on biological characteristic and to examine the expression of target gene. Also determine the adenovirus infection rates by using Ad-GFP as a control gene vector.4. To investigate the feasibility of using ADSCs as seeding cells and chitsan tricalcium phosphate (CTCP ) as a scaffold, and to observe the capability of fabricating the tissue engineering bone in vivo. Also observe the characteristic of ADSCs transfered with BMP-2 gene in immunity-dificency nude rats and SD rats...
