Objective: By applying the principle and technology of tissue engineering bone, the experiment aims to develop three-dimensional porous and double effective composite gradient scaffold material with PLA/chitosan/Collagen, and carry out external tissue engineering bone-cartilage construction and the research of induced regeneration of primary bone-cartilage in vivo.Methods: To develop three-dimensional porous composite gradient scaffold material with derivative of chitin /chitosan/collagen by using technology of biodegradation crosslinking agents and ion solvent and supercritical method; induce to transform externally rabbit marrow mesenchymal stem cells into cartilage cells, and construct cartilage composite with bone scaffold by jointly fostering with gradient material; transplant the composite into New Zealand rabbits for the regeneration of primary bone— cartilage and repair of articular cartilage defects. Results: 1, the strengthen of biomechanics, porosity and aperture of the developed gradient scaffold material met expected demands; 2, rabbit marrow mesenchymal stem cells were externally transformed into cartilage cells and successfully grew inside the material and lined up in one layer; 3, articular cartilage defects with a diameter of 4mm were basically repaired after transplanting the externally-constructed bone—cartilage composite into New Zealand rabbits.Conclusions: 1, the strengthen of biomechanics, porosity and aperture of the gradient material meet the demand of cartilage growth in situ and regeneration in vivo; 2, in the condition of induced and centrifugal cultivation, rabbit marrow mesenchymal stem cell can successfully grow cartilage tissue, and cartilage cells can grow in one layer in the material, preliminarily constructing cartilage cell composite with bone scaffold; 3, bone-cartilage composite can induce bone—cartilage regeneration and...
