Objective: To investigate the feasibility of Creating tissue engineering heart valve from bone nesenchymal stem cells as an alternative cell source and collagen-chistosan complex as a scaffold.Methods:To attain rabbit bone marrow under sterile conditions, isolate and purify BMSCs using adherent culturing method, in vitro culture and expand and induce to differentiate endothelial cells and assay/characterize; In vitro isolate, culture and expand rabbit aortic wall fibroblast and snooth muscle cells by tissue mass and assay; To isolate, culture and expand rabbit aortic wall fibroblast and smooth muscle cell and endothelial cell seeding on collagen-chitosan scaffold by premoisture treatment, in vitro cuturing and observing, at the same time, scaffold materials were implanted on surface of New Zealand white rabbit laissimus dorsi , taked them out and examined periodically endothelial cell adhesion , proliferation , growth, stroma secretion and tissue compatibility and biological degradation in vivo by histology, inverted microscopy, transmission electron microscopy.Results :1. Better hydrophilia and cell affinity of collagen-chitosan complex scaffold, subcutaneous experiment showed good biological compatibility, in vivo. degration time was between 8 and 10 weeks: 2. BMSCs grew by adherent wall and could be induced to differentiate into endothelial cells, Curve of cell growth was good. In vitro succeeded to isolate, culture and expand rabbit aortic wall fibroblast and smooth muscle cells by culture mass; 3. In vitro TEHV might be constructed using fibroblast, smooth muscle cell and endothelial cell as seeding cells and collagen-chitosan complex materials as scaffold: endothelial cells could adhere, extend, proliferate on collagen-chitosan scaffold and secreted stroma by HE...
