Impaired glucose tolerance during pregnancy is a problem faced by each pregnant woman , it will lead to various diseases of mothers and infants .Lots of researches have shown that insulin resistance is closely related to abnormal glucose metabolism in pregnant women.It has been proved that insulin receptor species (IRSs), phosphatidyl kinase, glucose transporters (GLUTs), and nuclear receptors are the key protein ( or enzymes) elements involved in participating and regulating glucose metabolism in cells.The specific topic through pregnancy trophoblast cell glucose metabolism study, TCM extracts - Berberine, bilingual saponins intervention , discuss glucose metabolism correlation protein expression change in trophoblast cell and control action of medicine ,simultaneously studies the influence of trophoblast cell glucose metabolism disorder on hormone secretion. Methods: the gene expression related molecules was detected by real time Polymerase chain reaction (RT-PCR); protein expression was detected by immune protein (Western blot); immune cells chemistry in combination with laser scanning technology for molecular and cellular localization; placental hormone levels in the use of RIA or chemiluminescence. The results : 1. With treatment of Wortmannin ,~3H-D-glucose incorporation reduced compared with normal P <0.05; Supernatant residual glucose content rised compared with normal P <0.05; P110-induced mRNA expression by RT-PCR decreased 90.2%. 2. With wortmannin blocking IRS-1, phosphatidylinositol 3 - kinase (P85) and PPARy mRNA expression reduced. 3. Berberine can significantly improve the IRS, P85, PPARγmRNA expression. Berberine is similar to Troglitazone on the regulation of IRS-1 and PPARy,and respectively increased by 23.94% and 113.9% with intervention; P85 mRNA elevated 64.41%, that obviously superior to other drugs; the regulations of diosgenin on P85 and PPARγwere up to a certain extent, and respectively 17.91% and 10.57%.Protein and mRNA expression had the same trend, but the level of regulation is not significant; IRS-1 distributed in the cytoplasm with medium expression. GLUT1 distributed in the cytoplasm and membrane with expression in less; PPargamma distributed in the nuclear membrane with lower expression. 4. MLN64mRNA expression : the expression in WT group reduced 26.91% (0.326/0.446) compared with normal cells, its expression in B, T group elevated significantly compared with the WT, respectively 69.93% (0.554/0.326) and 53.98% (0.502/0.326); Changes of placental hormone secretion: compared with the WT, E2 and P were significantly increased,P<0.05, the other drug had no significant effect on the secretion of hormones.Summing up results showed : 1. Wortmannin can make it abnormal the uptake and use of glucose in trophoblast cells to establish cell glucose metabolism disorders model. Trophoblast cells may exist with insulin signal transduction in cells in a similar way to participate glucose metabolism. 2. Berberine on various important role in regulation of glucose metabolism associated protein factor, this may be improve glucose metabolism disorder in the intracellular molecular mechanisms, and in some areas nurtured results is similar to troglitazone and metformin. 3. Diosgenin had no significant role in the regulation of glucose metabolism in trophoblast cells. 4. MLN64mRNA decreased where trophoblast cells glucose metabolism obstacle. It may affect limited-steps of synthetic steroid hormones . Berberine and Troglitazone can increase its expression, and a considerable role. 5. Other drugs in addition to troglitazone had not effect significantly on the placental hormone secretion.
