Objective: (1) To explore the expressions of Nuclear factor kB in the synovialis and spine ganglion of the adjuvant arthritis of rats (AA) , discuss the mechanism of the RA from nerve-incretion-immunology. (2) To explore the expressions of NF-kBmRNA and TNF-αmRNA, IL-1βmRNA, IL-10 mRNA in the synovialis, discuss their relationship and the levels of expressions of RA. (3) To explore the results confero of the routine acupuncture (RA) and acupuncture of Stimulating Du Channel and Smoothing Other Channels and Collaterals (ASDCSOCC) on the AA.Methods: Project four rat groups of blank control, model control, routine acupuncture (RA) and Acupuncture of Stimulating Du Channel and Smoothing Other Channels and Collaterals (ASDCSOCC) group, study on the AA. (1) Observe the changes of the arthron swelling, synovialis and spine ganglion. (3) Observe the changes of the synovialis ultrastructure by using the transmission electron microscope. (4) Detecting the expressions of Nuclear factor kB(P65)in the synovialis and spine ganglion by using the immunity organization chemistry (IOC). (5) Detecting the expressions levels of the TNF-αmRNA, IL-1βmRNA, IL-10 mRNA and NF-kBmRNA in the synovialis by using the reflection of the RT-PCR.Results: (1) The expressions levels of the TNF-αmRNA , IL-1βmRNA and NF-kBmRNA in the synovialis of the model control group raised clearly, and they had distinct disparity comparing with the blank control group (p<0.01). The expressions levels of the IL-10 mRNA of the model control group was descending, and it had distinct disparity comparing with the blank control group(p<0.05). After treating by the RA and ASDCSOCC, The expressions levels of the TNF-αmRNA , IL-1βmRNA and NF-kBmRNA are descending, and they had distinct disparity comparing with the model control group(p<0.05, p<0.01), but the expressions levels of the IL-10 mRNA of the ASDCSOCC group was raising , and it had distinct disparity comparing with the RA group(p<0.05). After treating, comparing with ASDCSOCC group and RA group had distinct disparity (p<0.01). (2) The relationship of the expressions levels of the TNF-αmRNA , IL-1βmRNA with the NF-kBmRNA are linear and positive (p<0.01), but the IL-10 mRNA is negative (p<0.01) . (3) The immunity organization chemistry staining of the synovialis and spine ganglion NF-kB (P65) display the masculine reactant of the NF-kB (P65) emboldening clearly, and form like the ball distribution, and the color of the nucleolus in the staining is deeper than in the cytoplasm. The model control group is higher than the blank control group, and they had distinct disparity (p<0.01). After treating by the RA and ASDCSOCC, comparing with the model control group they had distinct disparity (p<0.05, p<0.01) . Comparing with the ASDCSOCC group and the RA group, they had distinct disparity (p<0.05) . (4) The method of the HE staining observation, the pathology integration of the model control group is raised clearly comparing with the blank control group, and they had distinct disparity (p<0.01) . After treating, the pathology integration of the RA and ASDCSOCC are descending, comparing with the model control group they had distinct disparity (p<0.01) . Comparing with the RA group, the pathology integration of the ASDCSOCC group is lower, and they had distinct disparity (p<0.05). (5) Observing by the transmission electron microscope , the numbers of the chondriosome affection, swelling, carination affection and the degeneratio of the synovialis cell are raising clearly, comparing with the blank control group, and they had distinct disparity (p <0.01) . After treating by the RA and ASDCSOCC, all of the norm above had reduced, and they had distinct disparity comparing with the model control group (p<0.01). The ASDCSOCC and group and the RAgroup had distinct disparity (p<0.01). (6) Comparing with the blank control group, the level of the articulations swelling, flex articulations ache and arthrophlogosis of the model control group, and they had distinct disparity (p<0.05, p <0.01) . After treating by the RA and Acupuncture of Collaterals, all of the norm above had reduced, and they had distinct disparity comparing with the model control group(p<0.05, p<0.01). The ASDCSOCC group and the RA group had distinct disparity (p<0.05) .Conclusion: (1) The expressions of Nuclear factor kB in the synovialis and spine ganglion of the adjuvant arthritis of rats (AA) are raising , and the NF-kB joined in the etiopathogenesis of the whole body pathologic changed, they form the network of the nerve- immunity. (2) The relationship of the NF-kB and TNF-αIL-1βin the synovialis are linear and positive, they influence each other, and they influenced by the aggravation level of the ailment. (3)The level of the metatarsus swelling, arthrophlogosis and flex articulations ache of AA are reduced after treating by the RA and ASDCSOCC, and have the very clearly effect of antiphlogistic, apocatastasis and analgecize, and the method of Acupuncture of Stimulating Du Channel and Smoothing Other Channels and Collaterals better than the routine acupuncture. (4) The inflammatory cell infiltration, vein hyperplasia, the form of the pannus and the injury of the ultrastructure in the articulation synovialis telae of AA are reduced by treating of method of the RA and ASDCSOCC , so reduce the level of synovialis telae accrementition , articulation cartilage and bone injury and articulation swelling. And we find that the result of the ASDCSOCC is better than the RA. (5) The spine nerve pathology injury of the AA are reduced by treating of the method of the ASDCSOCC and, the nerve- immunity network is a one of the most important factors of the etiopathogenesis of RA, and the effect of the ASDCSOCC method is better than the RA. This is the one of the unbalance label of the nerve-immunity network by the acupuncture treating. (6) The expressions levels of the TNF-αmRNA and IL-1βmRNA in the synovialis telae are reduced and the IL-10mRNA is raised after treating by theRA and ASDCSOCC and, these are the one of the molecule mechanism of RA by acupuncture treating. And we find that the effect of ASDCSOCC is better than the RA. (7) The abnormality activation of NF-kB and abnormality expression of mRNA in the synovialis telae and spine ganglion are reduced after treating by the RA and ASDCSOCC. This is one of the gene transcribe and control level mechanisms, and the the effect of ASDCSOCC is much better than the RA. (8) It has very clearly adjustment effect of the never and the immunity disharmony of AA by the acupuncture, to cure the RA and reach to natural state of the Yin and Yang Equilibrium by the Strengthening Body Resistance to Eliminate Pathogen in the traditional Chinese medicine mechanism.
