On the beginning of the theories of AD, which were the hypothesis of nerves immune inflammation and the hypothesis ofβ-amyloid protein cascades, and based on that the metabolic pathway of Aβparticipated in AD, The topic adopted Aβ1-40 to inject the Hippocampus to induce neuronal damage in brain, to copy the AD rat model. By the curative target ofβ-amyloid protein deposition and neuronal damage pathological changes, we used Morris water maze and neuropathology to observe the changes of ethology and brain histomorphology of AD rat; used radioimmunity method to determine the levels of Aβ, IL-1β, IL-6 and TNF-αin serum and Hippocampus of rat; and used immunohistochemical method to detect the expression of Aβ1-40,β-APP, COX-2, nNOS and IKB-αin Hippocampus of rat; also used RT-PCR method to detect the expression ofβ-APPmRNA, IL-1βmRNA, IL-6mRNA and TNF-αmRNA in Hippocampus. Then we discussed the therapeutic efficacy and amynology mechanism of Buyang Huanwu Decoction for AD rat induced by Aβfrom the ethology, cell, molecule and gene levels, therefore provided the experimental evidences for clinical application.The results of the study indicated that Buyang Huanwu Decoction had obvious improved effects for dysmnesy of AD rat model induced by Aβ; could lower the obviously increased Aβdeposition markedly in Hippocampus and brain mantle; and improved neuronal damage caused by Aβ, and improved nerves fibronolysis damage markedly, lessened inflammatory reaction in brain; so could lower obviously the abnormal high expression ofβ-APP protein and mRNA in Hippocampus of AD rat caused by Aβ; lowered high levels of Aβ, IL-1β, IL-6 and TNF-αin serum and Hippocampus and the abnormal high expression of inflammatory cytokine IL-1β, IL-6 and TNF-αmRNA in Hippocampus of AD rat model; then steped up the masculine expression of nNOS in Hippocampus of AD rat model; finally could lower high masculine expression of COX-2 and IKB-αin Hippocampus. During the indicatrixes mentioned above, the effects of Buyang Huan wu Decoction group were most obviously, and the group of promoting blood flow surpassed that of invigorating vital energy.Buyang Huanwu Decoction might regulate immunity inflammatory cytokine IL-1β, IL-6 and TNF-αof blood and brain, also gene expression of that, and interfere in the generation and metabolism ofβ-APP, control reasonably the immune inflammation cascade reaction caused by Aβdeposition through signal transduction pathway of COX-2, IKB-αand nNOS. Thereby lessened brain neuronal damage effectually caused by Aβtoxic action, and improved the dysmnesy. Then achieved the effect of prevention and cure of AD.
