| Objective:To study the effect of Buyang Huanwu Decoction(补阳还五汤BYHwD)on the ultramicrostructure,expression of GAP-43 and Synapsins-1 in brain tissue and explore the possible mechanism involved.Method:80 rats were randomly divided into a sham operation group,a model group,a BYHWD group,and a ultramicro powder group,which were made ischemia—reperfusion injured with the method of MCAO.The brain tissue were isolated in different time and its pathomorphology and ultramicrostructure were observed by light microscope and electron microscope.The expressions of GAP-43 and Synapsins-1 in the spinal tissue were investigated with the immunohistochemical methods.Results:1.The expression of GAP-43 and Synapsins-1was of no change in the sham operation group,lower obviously from 7th day after ischemia—reperfusion injury in the model group,and higher in the BYHWD and ultramicro powder group from 14th day after ischemia—reperfusion injury(P＜0.01),compared with the sham operation group.2.It was observed by light microscope that there were no infarction in the sham operation group,and the infarction volume in other groups were (134.15±18.26)mm3 in the model group,(78.46±11.32)mm3 in the BYHWD group and(80.23±9.23)mm3 in ultramicro powder group,which was significantly lower than the model group.3.It was Observation by electron microscope that the ultramicro-morph of cells in the sham operation group was normal,oedema,vacuole and apoptotic body were visible in the nerve cells and glial cells in the model group,and those were partly seen in the BYHWD and ultramicro powder group.Conclusion:BYHWD can reduce the infarction volume,protect the nerve cells and improve the nerval founction after spinal ischemia-reperfusion injury.The mechanism of this maybe that BYHWD up-regulates the expression of GAP-43 and Synapsins-1.The effect of ultramicro powder is the same with the BYHWD,but its dosis is only half of the BYHWD.That is to say the ultramicro powder outweighs the BYHWD in the efficacy and material-saving.
