The Experimental Study Of The Effects Of Yangshenyutangwan On The Expression Of TNF-α,IL-6 And GLUT4 Etc. Of STZ-2DM Rats

Objective:to investigate the action and mechanism of Yangshen Yutang Wan on how to influence type 2 diabetic rat's blood glucose, blood lipid,INS,Leptin,HDL-C,LDL-C,GLUT-4 mRNA,RAGE mRNA,MMP-9mRNA, iNOS mRNA and pathologic changes, and improve their disorder state of the Wistar rats models by injecting STZ in the abdomen.Methods: Establish 2DM rats models by intraperitoneal injection of STZ and divide randomly in six groups, ie. model control group, low dosage of Yangshen Yutang Wan, middle dosage of Yangshen Yutang Wan, high dosage of Yang-shen Yutang Wan, Metformin Hydrochloride group, Tang-niaole group respectively. Normal control group was also established. After four week's treatment, detecting rat's following parameters including BW, FPG, FINS, HDL-C, LDL-C, meanwhile rat's pancreatic tissues are also made into pathological section for analysis. and rat's muscles, fat tissues made analysis mRNA of GLUT-4,RAGE,MMP-9,iNOS. The effect of different medicines on the these parameters were detected by radiommunoassay,Biohemistry method,Histomorphometry,RT-PCR, etc.Result:2DM rats do exist abnormality of the above mentioned index,whose rising blood lipid level is one of important lipid metabolic disorder. Yangshen Yutang Wan can improve the DM rat's blood glucose and blood lipid metabolic disorder and can reduce INS in blood serum, TNF-α,IL-6, meanwhile increase HDL-C content in blood serum whereas descend LDL-C, accelerating the repair and protection of the impaired pancreatic cell. Compared with normal control group, the expression of RAGEmRNA, MMP-9mRNA and iNOSmRNA increased whereas the expression of GLUT-4mRNA reduced in model control group. Compared with model control group, the expression of iNOSmRNA,RAGEmRNA and MMP-9mRNA reduced in low dosage,middle dosage and high dosage of Yangshen Yutang Wan groups. Compared with model control group the expression of GLUT-4 increase in low dosage,middle dosage and high dosage of Yangshen Yutang Wan groups. The general curative effect of Yangshen Yutang Wan is better than the west medicine cross-refer group.Conclusion: It has been successful to establish diabetes rats model by 30mk/kg dosage STZ that was injected. Yangshen Yutang Wan is an efficacious prescription for Diabetes Mellitus.
