Study On The Mechanism Of The Treatment Of Insomnia By Acupuncture At Five Zang-Organs And Regulation At Five Zang-Soul

ObjectiveTo heal insomnia based on adjusting five viscera according to clinical study,To study the pathological experimental and discuss the pathogenesis through insomnia's rats with PCPA in order to supply the basic theory and experimental data for prevention and treatment of insomnia by acupuncture at five zang -organs and regulation at five zang-soul, further explain theoretical connotation on five zang -soul,psychology and somnus.Methods1 .The rats model was induced by PCPA peritoneal injection.2.The 5-HT, 5-HIAA, NE, IL-1βTNF, GABA, GABAA and PGE2 in brain tissue was measured by euzymelinked immunosorbent assay and RI.3.At random, the mice were divided into 4 groups: normal group,model group, diazepam group , acupuncture group. We examined the 5-HT, 5-HIAA, NE, IL-1βTNF, GABA, GABA_A and PGE2 in brain tissue.4.Spss 11.0 was used. Data was analyzed using one-way ANOVA. All values were expressed as mean±SE,statistical significance was assigned at a P value of <0.05 or <0.01.Results1.Normal sleeping cycle of rats - awareness disappearec by PCPA peritoneal injection, it ' s a good model of copying insomnia .2.Acupuncture could coordinate relations of transmitter, advance 5—HT and 5-HIAA, degrade NE, the difference was significant comparing model group (P<0.05) .3.Acupuncture could increase IL-1βand TNF,the difference was significant comparing model group (P<0.05) .4.Acupuncture could increase GABAand GABAA,the difference was significant comparing model group (P<0.05) .5.Acupuncture could depress PGE2,the difference was significant comparing model group (P<0.05) .Conclusions1 .Acupuncture five viscera point could increase 5—HT and 5-HIAA in hypothalamus of rats, achieve normal level,on the other hand, obviously decrease NE. It indicated the acupuncture could coordinate relation of transmitter, advance 5—HT, 5-HIAA and NE.,then recovere normal rule of sleep - awareness, achieving to treat insomnia.2.Acupuncture five viscera point could increase IL-1βand TNF in hypothalamus of rats, achieve or exceed normal level. It indicated the acupuncture could adjust immune function and increase the cytokine of promoting sleep.3.Acupuncture five viscera point could increase GABA and GABA_A,almost achieve normal level, It indicated the acupuncture could increase slow-wave-sleep4.Acupuncture five viscera point could obviously decrease PGE2 in hypothalamus of rats, almost achieve normal level. It indicated the acupuncture could decrease awareness, increase SWS and REMS.5.Diazepam could recover PCPA rats's cycle of sleep -awareness, obviously improve the indicatrix of experiment. The difference was quiet (P>0.05) in acupuncture and diazepam group, but experiment indicated acupuncture group a little surpassed diazepam group.
