Objective: Two-dimension electrophoresis integrated with massspectrometry, which has made rapid development in the area ofproteomics, is one of the most major new methods for protein analysis. Inthis study we aim at making the protein database of normal endometriumby two-dimension electrophoresis and mass spectrometry.Methods: Two-dimension electrophoresis combined with massspectrometry was used for normal proliferative endometrium, originatedfrom two young, married and healthy women. Two-dimensionelectrophoresis combined with mass spectrometry was also used fornormal proliferative endometrium from 3 control healthy young women,eutopic proliferative endometrium and endimetrioma wall from 3 cases ofendometriosis.Result: High-qualified two-dimension electrophoresis picture, consistingof hundreds of protein spots, was completed. We found 113 differentvolume spots between normal endometrium and endometrioma wall, 44different volume spots between normal endometrium and eutopicendometrium of endometriosis patients, 164 different volume spotsbetween eutopic endometrium and endometrioma wall of endometriosis patients. There were 10 different protein be identification and we tooktwo of them, Prohibitin and hnRNP A2/B1 to further reseaching..Conelusion: Two-dimension electrophoresis, preparing for massspectrometry, is a highly sensitive technique to separate the tissueproteins from endometrium. There were many differents among normalendometrium, eutopic endometrium and endimetrioma of endometriosis. Objective: To evaluate the expression and localization of hnRNP A2/B1and Prohibitin in endometriosis.Methods: Samples were obtained from three normal proliferate-stageendometria of young married and healthy women, three ovarianendometriosis and three eutopic endometria of three patients. Theexpression of Prohibitin and hnRNPA2/B1 was examined byimmunohistochemistry and Western blot with a specific polyclonalantibody.Result: Prohibitin was located in the cytolymph of both epithelial glandsand stroma; hnRNP A2/B1 was located in the nucleus of both epithelialglands and stroma. The expression level of these two proteins in theendometriotic tissues and the eutopic endometria of patients withendometriosis were both weaker than those in the normal endometria.The difference between the endometriotic tissues and eutopic endometria wereboth insignificant.Conclusion: Prohibitin and hnRNP A2/B1 may play important roles inthe pathogenesis of endometriosis.
