Blumea riparia DC. (Compositae) has been used as a folk herbal medicine to treat the gynecological diseases, such as menorrhagia, puerperal metrorrhagia, puerperal edema, FUB. barrenness, yin sore, for a long time in Guangxi Province, China.The chemical constituents of Blumea riparia DC. were studied by multiple chromatographical techniques (such as silica gel chromatography, sephadex LH-20, PTLC and HPLC etc.). On the basis of physicochemical evidence and spectral analysis, the structures of 48 compounds obtained from its 70% ethanol extract were identified as follow: Blumeaxanthene(1), Blumeacetylene A (2), Blumeacetylene B (3), ichthyothereol (4). Blumeaguaianone A (5), Blumeaguaianone B (6), Blumeaguaianone C (7), 1-angeloyloxyeudesm-4, 7-diol (8), (+) lα, 9β-dihydroxy-7α-H-eudesm-4-en-6-one (9), cryptomeridiol (10), (+)dehydrovomifoliol (11), (-) 3β-hydroxy-5α, 6α-epoxy-7-megastigmen-9-one (12), (-) loliolide (13), austroinulin (14),β-sitosterol (15), stigmasterol (16), (24 S) stigmast-4-en-3-one (17), (24 S) stigmast-4, 22-dien-3-one (18), taxifolin-4'-methyl ether (19), eriodictyoi-7, 4'-dimethyl ether (20), eriodictyol-7, 3'-dimethylether (21), eriodictyol-7-methyl ether (22), quercetin-7,3',4'-trimethyl ether (23), quercetin-3,7,4'-trimethyl ether (24), tamarixetin (25), rhamnocitrin (26), kaempferol (27), quercetin-7,4'-dimethyl ether (28), quercetin-7-methyl ether (29), quercetin (30), quercetin-3,7,3',4'-tetramethyl ether (31), syringaresinol (32), hydroquinone (33), protocatechuic aldehyde (34), 2-hydroxy-4, 6-dimethoxy-acetophenone (35), 2,4-dihydroxy-6-methoxy-acetophenone (36), 4-hydroxybenzaldehyde (37), benzyl-O-β-D-glucopyranoside (38), 3-(3,4-dimethoxyphenyl) propanamide (39), protocatechuic acid (40), caffeic acid (41), 2, 4-dimethoxy-6-methyibenzoic acid (42), syringic acid (43), gallic acid (44), vanillic acid (45), furoic acid (46), salicylic acid (47), cyclo-(l-pro-l-leu) (48).Among them, 8 compounds are new compounds. Compound 1 is a new xanthene, Compounds 2, 3 are two new polyacetylenes, Compounds 5, 6, 7 are three new sesquiterpenes with the skeleton of guaian-en-one. Compounds 8, 9 are two new eudesmane-type sesquiterpenes. The compounds obtained are isolated from this plant for the first time except 34 and 40.The cytotoxic activities in vitro of some compounds have been assayed against three cancer cells lines: HL-60, U937 and T47D. Among the compounds evaluated, compound 1 showed strong activities against the cancer cells, HL-60 with IC50 value of 7.45μmol/L.
