Objective: This paper took Blumea riparia(Bl.)DC.as research object.The spectrum-effect relationship of its hemostatic effect is studied,and the quantitative analysis multi-components by single marker(QAMS)mode of Blumea riparia(Bl.)DC.is established to provide basis and reference for the quality improvement of Blumea riparia(Bl.)DC..Methods: The samples of Blumea riparia(Bl.)DC.from 20 different areas are taken as research objects.Using HPLC fingerprint technology is selected to research the fingerprint of Blumea riparia(Bl.)DC.,to establish chromatographic conditions,implementing methodological study.Carriying out similarity analysis,cluster analysis and principal component analysis of the fingerprint to mine the chemical information of composition peaks of Blumea riparia(Bl.)DC..Using the warfarin model in the study of pharmacodynamics part,taking blood coagulation of the four indicators(PT?APTT?TT?FIB)as index to study the hemostatic activity of Blumea riparia(Bl.)DC.of different areas.Using Grey Relational Analysis and associating information of each standardized common chromatographic peaks with pharmacodynamics data to screen chromatographic peaks,which are closely related to the hemostasis effect.On the basis of fingerprint peak information and pharmacodynamics data of 15 samples,using BP artificial neural network technology to establish fingerprint-efficacy relational model.The remaining 5batches of sample data are used to verify the accuracy and reliability of the model.With chlorogenic acid as internal standard,using QAMS technology to calculate the relative correction factors of protocatechuic acid,caffeic acidand rutin.Determining the content of each component,and comparing the results with external standard method to evaluating the accuracy of QAMS model.Results: The common model of Blumea riparia(Bl.)DC.fingerprint is stable and reliable.A total of 13 common peaks are identified: the No.2 peak is protocatechuic acid,the No.3 peak is chlorogenic acid,the No.5 is caffeic acid and the No.6 is rutin.The similarity degrees of 20 samples of Blumea riparia(Bl.)DC.are more than 0.7,which means the components are stable.20 samples of medicinal materials are divided into 3 major categories by cluster analysis.S-10 and S-12 belong to the first kind.S-1,S-2,S-3,S-5,S-6,S-7,S-8,S-9,S-11,S-13,S-14,S-15,S-19 and S-20 belong to the second kind.S-4,S-16,S-17 and S-18 belong to the third kind.The differences in classification are mainly on the No.2,No.3,No.5,No.9 and No.10 peak.The principal component analysis obtained the peak numbers of the variables with maximum weight value are No.3,No.10,No.9,No.2,No.7 as first 5,and the weight values are 0.969,0.929,0.919,0.905,0.841.The results of two analyses all show that No.2,No.3,No.9,No.10 peak have important influence on the quality of medicinal materials.The results of pharmacodynamics study show that Blumea riparia(Bl.)DC.has hemostatic efficacy.20 samples of Blumea riparia(Bl.)DC.show different degrees of hemostatic effect on warfarin rats.The results show that it can reduce PT,APTT and TT values of rats significantly,and bring the FIB value an increasing trend.The analysis of grey correlation degree shows that correlation coefficient of 13 common peaks with pharmacodynamics are above 0.7.The characteristic peaks of established fingerprint show positive correlation with hemostatic efficacy.Protocatechuic acid,chlorogenic acid,caffeic acid,rutin and some undetermined compound are the foundation of hemostatic activity of Blumea riparia(Bl.)DC.,and they have great association with its efficacy.Theestablished hemostatic spectrum effect relationship model of Blumea riparia(Bl.)DC.has good predictive performance for hemostatic efficacy.The absolute value of relative error of pharmacodynamics predictive results is less than or equal to 13.60%,and the relative error of most values is less than6.7%.The QAMS model of the Blumea riparia(Bl.)DC is established.The relative correction factors of caffeic acid and protocatechuic acid rutin are1.25,1.88 and 0.40,respectively.And the RSD are 0.29%,0.38% and 0.33%.The relative correction factor of the components has good reproducibility and stability.The correlation coefficient results of QAMS and external standard method are greater than 0.999.The relative error is less than 3%,which means there is no significant difference between the results of these two methods.Conclusion: The research of spectrum-effect relationship of Blumea riparia(Bl.)DC.and the study of chemical components content establish a "component"-"peak"-"effect"-"quantity"-"quality" evaluation system of Blumea riparia(Bl.)DC..The quality of medicinal material of Blumea riparia(Bl.)DC.can be measured in three dimensions.The study also provides valuable experimental basis for the further development and utilization of Blumea riparia(Bl.)DC..
