Phellinu linteus (PL), an orange color mushroom growing well on mulberry tree, is awell-known fongus of the genus Phellinus in the family of Hymenochaetaceae and has beenused as a traditional herb medicine in oriental countries. To study on effect of Phellinuslinteus on the anti-tumor activity and immunomodulatory action of mice beating tumor.Experiment includes four aspect. First, we study the anti-tumor activity of Phellinuslinteus, second, we study on efficacy enhancing and side-effects reducing ofPhellinus linteus on radiotherapy or chemotherapy, third, on effect of Phellinus linteus onimmunomodulatory action of mice bearing tumor its, and fourth, a mechanism of thatPhellinus Iinteus adjust immune function of mice bearing tumor.First,we study the anti-tumor activity of Phellinus linteus. Solid tumor was induced byinjecting MFC, HepA, S180 and Lewis cells to study on effect of high-dose, mean-dose andlow-dose Phellinus linteus (2.0g/kg, 1.0g/kg, 0.5g/kg)on the anti-tumor activity in vivo.Result suggests that all three dose group of Phellinus linteus had inhibited tumor weight ofmice bearing MFC, HepA tumor cell (P<0.05, P<0.01). Group of high-dose and mean-dosePhellinus linteus had inhibited tumor weight of mice bearing S180 tumor cell(P<0.05, P<0.01).Group of high-dose Phellinus linteus had inhibited tumor weight of mice bearing Lewis tumorcell (P<0.05), but had not function of anti-metastasis. Above-mentioned experiment do threetime again. Experiment in vitro suggest that Phellinus linteus can not kill straightly MFCtumor cell.Second, we study on efficacy enhancing and side-effects reducing of Phellinuslinteus on radiotherapy or chemotherapy. To study on efficacy enhancing andside-effects reducing of Phellinus linteus on radiotherapy or chemotherapy by theexperiment that PL with radiotherapy or chemotherapy treats mice implanted tumor. Micewere implanted MFC subcutaneously to the right anterior limb and separated randomly tothirteen groups: control, groups of CTX (20 mg/kg,10 mg/kg and 5 mg/kg) and groups ofPL (2.0g/kg, 1.0g/kg, 0.5g/kg) with CTX (20 mg/kg,10 mg/kg and 5 mg/kg) respectively. Miceof control were administered intragastricly at a dose 0.01 mL/g for 10 days; PL were administered intragastricly for 10 days; CTX were administered parenterally every other day for 10 days. The body weight and the tumor weight of mice were measured. Mice were implanted HepA subcutaneously to the lateral side of tibia of the right posterior limb and separated randomly to five groups: control, group of radiotherapy and groups of PL (2.0g/kg, 1.0g/kg, 0.5g/kg) with radiotherapy respectively. Mice of control were administered intragastricly at a dose 0.01 mL/g for 14 days; PL were administered intragastricly for 14 days; after PL were administered for 5 days, X-rays were utilized at a dose 0.5 Gy for 5 days. The body weight and the tumor weight of mice were measured. Results: Vs. control group, all groups of CTX and groups of PL with CTX respectively can decrease a tumor weight(P<0.01). Vs. the groups of the equal-dose CTX, all groups of PL with CTX respectively can also decrease a tumor weight(P<0.05 or P<0.01). Vs. control group, groups of CTX can decrease the body weight of the mice(P<0.05 or P<0.01),but the body weight of groups of PL with CTX respectively don't change remarkably. Vs. control group, all group of radiotherapy and groups of PL with radiotherapy respectively can decrease a tumor weight(P<0.01). Vs. group of radiotherapy, all groups of PL with radiotherapy respectively can decrease tumor weight(P<0.05 or P<0.01). Vs. control group, group of radiotherapy can decrease the body weight of the mice(P<0.05),but the body weight of groups of PL with radiotherapy respectively don't change remarkably.Third, on effect of Phellinus linteus on immunomodulatory action of mice bearing tumor its. The life span, thymus weight and spleen weight were measured after mice were executed. Phagocytosis of mice was detected using the method that macrophages engulfed neutral red. The effects of PL on lymphocyte proliferatlon, mitogen-lnduced proliferation of ICR mice splenocytes were measured by MTT. Meanwhile, lymphocytes were prepared as described above. Spleen cells suspension was used as the effect cells and L929 cells was used as the target cells. L929 cells engulfing neutral red determined the cytotoxic activity of NK. SRBC was injected to abdominal cavity to elevate the level of antibody production by B cells. Results suggest high-dose, mean-dose and low-dose Phellinus linteus and CTX increase life span of mice bearing EAC tumor cells(P<0.001). Index of the spleens and thymus mice bearing tumor of high-dose and mean-dose of Phellinus linteus were greater than those in positive control group(P<0.05), and can increase the activity of macrophage (P<0.05, P<0.01). The average absorbance of ly mphocytes in high-dose and mean-dose group was higher than that in the untreated control group. High-dose of PL can enhance the cytotoxic activity of NK cells(p<0.01).The numbers of antibody forming cells were increased by PL treatment at high-dose and mean-dose group(p<0.05, p<0.001).Fourth, a mechanism of that Phellinus linteus adjust immune function of mice bearing tumor. We determined the content of IFN-γ, IL-2, IL-6 and IL-10 from the serum of mice transplanted Lewis lung carcinoma by ELISA method. We observed the change of spleen shape by HE-stained method. We determined the content of CD4+ and CD8+ from the serum and spleen by Flow cytometry method. We determined the content of mRNA of IFN-7and IL-10 from the spleen lymphocyte by RT-PCR method. The results showed that the content of IFN-γand IL-2 were elevated by the PL at the three doses group (P<0.05, P<0.01, P<0.001). Whereas, the content of IL-6 and IL-10 the PL of the high dose and mean dose were decreased compared with control group(p<0.05, P<0.01). Germinal center of spleen mice teated by the PL at the three doses group enhance and cells hyperplasia of germinal center active(P<0.05, P<0.01, P<0.001). PL of the high dose and mean dose can raise the content of CD4+ from the serum and spleen(P<0.05), CD8+ had not change obviously. PL of the high dose and mean dose can increase the content of mRNA of IFN-γbut decrease IL-10 from the spleen lymphocyte.Above-mentioned suggests PL has anti-tumor activity and immunomnodulational effect and probably inhibit tumor by means of activating the immunomoduletence of the body.
