Objective: To discuss the effect of the proliferation and differentiation of rabbit BMSCs induced by the nonparticles for rhBMP-2 in 3-D culture in vitro. Methods: BMSCs are isolated from the bone marrow by fulling combined with adhering to culture plastic in different time;the 3-D culture system of BMSCs-PLLA/β-TCP is constructed;no- nparticles for rhBMP-2 prepared by the solvent evaporation with double emulsion technique;BMSCs influenced by nonparticles for rhBMP-2 in 3-D in vitro studied by the control group of rhBMP-2 adminstered only. Results: The method of fulling combined with adhering to culture plastic in different time isolating BMSCs from bone marrow is simple and easy;furthermore, the homozygosis of BMSCs is good and the injury of BMSCs is very little;BMSCs are compatible to grow, proliferate and differentiate in 3-D system of PLLA/β-TCP; Nonparticles for rhBMP-2 could induce BMSCs resulting in the more persisting and powerful effect of the proliferation, differentiation and bone formation in 3-D system in vitro. Conclusions: our experimental study shows that the fulling combined with adhering to culture plastic in different time, the 3-D culture system of PLLA/β-TCP and the nonparticles(PLGA) for rhBMP-2 are the optimal methods and techniques for the construction of the bone tissue engineering.
