Objective To investigate the mechanisms that chitosan prevents postoperative adhesion, and the ability that the chitosan hydrogel enhances wounds healing. Methods It was observed that chitosan inhibited the proliferation of fibroblasts and induced its apoptosis, then it was explored with the method of proteomics that chitosan influenced the expression of proteins of fibroblasts. And It was observed that chitosan hydrogel effected the healing velocity and quality of cutaneous wounds in normal rats and delayed wounds in diabetic rats. Results Chitosan could inhibit the proliferation of fibroblasts and induce its apoptosis, and the protein profile of fibroblasts has changed under the effect of chitosan. After the chitosan hydrogle was used, wounds healing of norman rats and diabetic rats were accelerated and their healing quality were improved. Conclusions Inhibitive effect of chitosan on the proliferation of fibroblasts and its induced effect on the apoptosis of fibroblasts can be one of mechanisms that chitosan prevents postoperative adhesion. And chitosan hydrogel can enhance wounds healing of rats.
