In this paper, on the basis of chemical and pharmacological studies, the processing principle ofPsoralea corylifolia L.was discussed.By these two methods of TLCS and HPLC. By TLCS,there's no differences between samplesprocessed with salt and Leigong's methods (P>0.05) in the content ofpsorlan, the raw samples andsamples processed by stir-fried (P>0.05) in the content of isopsorlan, but there's a greatdifferences between the samples processed by stir-fried and oothers (P<0.05). By HPLC, there's nodifferences between samples processed with salt and Leigong's methods (P>0.05 ) in the content ofpsorlan, there's no differences between samples stir-fried and oothers (P>0.05) in the content ofisopsorlan.For the first time, in the content determination of coumarins in Psoralea corylifolia L. byHPLC, mathematical model of relative retention time and relative retention peak area wasestablished. By comparison of model, the processing methods which could increase the coumarincontent were established. In the same relative retention time, the samples processed with Leigong'smethods had the largest relative peak area. Compared with raw samples, the samples processed withsalt was less in relative peak area when the relative retention time is less than 2, but the result isopposite when the relative retention time is more than 2. The relative peak areas of samplesprocessed by stir-fired simply were all less than raw samples.By GC-MC, the relative contents of bakuchiol in different processing samples were analyzed.After being processed, the contents of bakuchiol all decreased, the contents in the samplesprocessed with Leigong's methods decreased processed with Leigong's methods decreased 1/4.By ICP-AES, the trace elements which had relation ship with contents of kidney weredetermined. After being processed, the total content of Mg, Ca, Fe, Zn changed a lot, while thecontents of Mn and Cu haven't obvious change. The contents of Cu, Mn, Ca, Mg, Fe, Zn in theEtOH extract of raw samples and samples processed with Leigong's methods are 1/30~1/4 of totalcontent. In the view of organic compounds and inorganic ion content, the necessarity of processingand difference of different processing methods were veritificated.Pharmacodynamic studies had showed that the processing methods could affected the efficacyofPsoralea corylifolia L.After being processed, the actions of rasing leukocytes were enhanced inthe mice with leucopenia induced by cyclophosphamide. The action of increasing the weight oftesticle and preputial glands in samples processed with salt, samples processed with Leigong'smethod was 2 times of the actions in raw samples and samples processed by stir-fried simply, whileno obvious changes were observed in the weight of prostate. These results showed that the mainefficacies of Psoralea corylifolia L. were enhanced by processed with salt and processed withLeigong's method, the results also indicated that psorlan and isopsorlan are the main activecomponents.For the first time, the effect of anti protein viral was observed in Psoralea corylifolia L., whichshowed that the extract ofPsoralea corylifolia L. contain the active components of anti protein viral.But the efficiency wasn't obvious. The sample extracted with EtOH and dissolved with ester onlymade Sup35p unstable, with the existence of GuHC1, sample 15 extracted with EtOH and dissolvedwith ester can cure 30% yeast protein viral.Microstructure and spectroscopy of raw samples and different processed samples were studied,and for the first time, the index of powder character in the adulterant of Psoralea corylifolia L. was established.There was no obvious difference and no characteristic changes in the microstructure of rawsamples and samples processed with salt; but obvious difference in the microstructttre of rawsample and its adulterant was observed.For the first time, the IR spectra of raw samples and different processed samples were studiedin 2+n dimensional space. All the common peak ratio of raw samples and different processedsamples are less than 70%, while all the common peak ratios of different processed samples aremore than 70%. In HPLC finger print, the similarity of raw samples and processed samples are lessthan 0.8, the similarity of different processed samples are 0.83~0.88.The best processing technology was optimized by uniform design when processed under thefollowing conditions: soaked in 20% salt solution for 20%, then heated by strong microwave for210s, by determining the content of psorlan and isopsorlan and extraction ratio, the absolute error ofmodel value and predicated value was 6%~0.3%, the relative error of proven value and predicatedvalue of model was 1.8%~0.6%.In this paper, the processing principle of Psoralea corylifolia L. were studied by thecomprehensive application of CMM identification, CMM analysis, CMM pharmacology, CMMpreparation, molecular biology, statistics, computer technology and so on, under the guidance ofTCM theory and clinic experience. The result indicated that the key factors of"decreasing toxicityand increasing efficacy" in the processing of Psoralea corylifolia L. are "soaking" and "heating".The researching methods are systematical all round and feasible, providing a new thinking way forthe study of processing.
