Objective The purpose of our study was to investigate the effect of PTH on the expression of CTGF in human renal proximal tubular epithelial cell line HK-2, determine the signaling pathway which activated and investigate the effect of Genistein on the expression of connective tissue growth factor in HK-2 stimulated by PTH and further the mechanism. Method The expression of CTGF, the machnism by which PTH or Gen acted in HK-2 cells and phenotypic transformation of HK-2 cells was measured by real time-polymerase chain reaction (RT-PCR), western blot, EMSA and light microscopy. Results 1 .PTH promoted CTGFmRNA and protein expression in HK-2 cells with the maximal response of PTH at a concentration of 10"10M and the best stimulating time at 48 hour. 2. PTH increased CTGF expression through EGFR ,MAPK and NF-kB signaling pathway. PTH stimulated ERK phosphorylation depending on three signal transduction pathways: PKA, PKC and PI-3K. 3. Gen inhibited CTGF expression which PTH induced by bloking MAPK and NF-kB signaling pathway. Conclusion l.PTH induced CTGF expression increased in HK-2 cells and transdifferentiation of HK-2 cells. 2. PTH acted through EGFR ,MAPK and NF-kB signaling pathway.3.Gen inhibited the effect of PTH.
