Lymphocytes are deeply involved in the initiation and perpetuation of inflammatory response in inflammatory bowel disease (IBD). We investigated the altered protein expression profile of lymphocytes in TNBS-induced colitis. Moreover the role of p38 MAPK pathway in T lymphocyte immune response after TNBS- induced colitis was studied. In addition, we describe the oral efficacy profile and mechanism of action of active composition of the Chinese Herbal Compound Changtai granule (CT) in rodent TNBS colitis using Semi-quantitative RT-PCR, ELISA and flow-cytometry (FCM) analysis.The results showed that the altered proteins, including regulators of the cell cycle and cell proliferation, signal transduction factors, inflammatory factors, apoptosis-related proteins, and metabolic enzymes were observed in antigen-specific lymphocyte activation of TNBS-induced colitis. Particularly some apoptosis-related proteins differentially expression may be involved in the pathogenesis of colitis. P38 MAPK pathway was involved in the T cell immune response after TNBS-induced colitis and regulated the expressiom of cytokines, which play an important role in the pathogenesis of IBD. Moreover, the active composition of Changtai granule can regulate and modificate the proportion of T cell subsets, decrease mRNA level of several inflammatory mediators and cytokines release. So CT may be an effective treatment approach for inflammatory bowel disease for the potential anti- inflammatory and immunomodulatory effect of CT in TNBS colitis.
