Objective:To observe the effect of Compound ZiGui Cream (CZGC) in rat cutaneous incised wound healing and investigate the mechanism of accelerating the rat back wound healing.Methods:two full thickness dermal excision wound models with the diameter of 1.8cm were established on the back of 208 healthy male or fermal Wistar rat, which were randomly divided into5groups:model group, Jing wanhong goup(Control goup), the high dose of CZGC goup((CZGC-H), the medium dose of CZGC(CZGC-M)goup and the low dose of CZGC(CZGC-L) goup,40 rats per group.With 8 rats as normal control and Jingwanhong as Positive compare medicine.The rats in each group were extemal applied with corresponding drug,0.1 g each wound surface and 2 times a day. The normal control group without any drug. tissues were removed 3,7,10andl4 days after surgery. The sample was handled and embed with paraffin, HE stained to observe the skin regrowth state and observation of histological changes, new capillaries stereology changes.The wound surface,healing time and modeling scar area after 30days were observed and recorded, The expression of macrophages CD68 was detected by immunohistochemical method (SP method) in the granulation tissue dynamicly; The expressiom of bcl-2, bax protein was detected by the method of immunohistochemistry (SAP-APAAP))and the expressiom of VEGFmRNA was by RT-PCR. images of each group was analyzed by Image-Pro-Plus image analysis software. All counted data was expressed in the form of x±s, and dealed by SPSS16.0 statistics software. Results:The wound healing rate, wound healing time, wound granulation tissue thickness, granulation tissue capillaries stereological changes in scar size, the scar area after 30days and other indicators of the CZGC were significantly heighten than Model group(P<0.05 or P<0.01) in each time points of observation; compared with model group, the expression of CD68 and VEGFmRNA in the CZGC group increased significantly (P<0.05 or P<0.01). compared with model group, The expression of Bcl-2 of the CZGC group were higher than the model group in the 3,7,10 days, with statistical significance (P<0.05或p<0.01). CZGC of high dose at 7 days, medium dose at 7,14 days reduce the level of Bax expression, there are statistically significant (P<0.05), in addition to low dose of the CZGC, in almost all observation time points the ratio of Bcl-2/Bax was higher than the model group, with statistical significance (P<0.05或p<0.01).Conelusion: The CZGC can promote wound healing in rat skin incision, reduce the scar size and improve the quality of wound healing.its mechanism of accelerating skin wound healing Possibly relate with an enhanced local immune and the expression of VEGFmRNA in granulation tissue so as to promote the proliferation of new capillaries, as well as inhibiting the process of wound healing apoptosis and other factors.
