ROLE OF HRG/ ErbB SIGNAL SYSTEM IN THE PROLIFERATION OF 22RV1PROSTATE CANCER CELL LINEObjective:Investigating the mechanism of prolilferation of 22RV1 cell inducted bythe signal of HRG/ ErbB system.Methods: First,detecting the expression of HER-2 and HER-3 by the confocalmicroscopy and flow cytometry;Secend,using the methords of MTT assay to examining theinfluence of HRG on 22RV1 cell prolilferation;Third, using the technology of flowcytometry and westernblot to investigate the mechanism of prolilferation of 22RV1 cellinducted by the HRG/ ErbB signal system.Results: 1.The FITC conjugated HER2 and PE conjugated HER3 are low levelespressed on 22RV1 cells. 2. FITC-HER2%﹦(6.4800±2.39729)%,PE-HER3%﹦(4.6200±1.33492)%. 3. HRGβ1 promtote the proliferation process of 22RV1 cells. 4.HRGβ1 activat the passway of PI3K/AKT. 5. Under the interfere of HRGβ1 , the cellPercentage of S phase become increased followed by the decrease of G0/G1 phases cellPercentage. 6. With the prolong of the HRGβ1 intervention, it also happens the increasingresult of cyclin D1 expression. 7. HRGβ1 can active NF-κB passway in a quickly manner.Conclusion: The HRG/ ErbB signal system can promtote the proliferation processof 22RV1 prostate cancer cells, the mechanism involves in is activating of the PI3K/AKTpassway and the increasing expression of cyclin D1 also with the activating of NF-κBpassway.
