Background and Objectives:To study siRNA interfere the expression of Bcl-2 mRNA investigate the effect of siRNA-bcl-2 on proliferation,apoptosis and sensitivity to chemotherapy of childhood B-ALL.Methods:36 children patients with newly diagnosed B-ALL were selected, so as to control group. Bcl-2 expression were assayed and compared between B-ALL children and control group by real-time RT-PCR. The bcl-2-siRNA was transfected into leukemia cells of chemotherapy resistant group by liposome and Cell proliferation and apoptosis were detected by MTT method. The chemotherapy resistant group leukemia cells were treated with both bcl-2-siRNA and daunorubicin and cell proliferation were detected, and the clinical application prospect of si RNA in children with B-ALL was analysis.Results:①Bcl-2 expression in BMMCs of newly diagnosed childhood B-ALL was 6.24±0.87, significantly higher than that in the control group(t=3.82, p<0.05). Bcl-2 expression of High risk group was 7.71±0.34, significantly higher than that in standard risk group(t=2.91, p<0.05).②Bcl-2 expression in chemotherapy resistance group before chemotherapy was 7.26±0.24,much higher than that in chemotherapy sensitivity group(t=3.07, p<0.05). Bcl-2 mRNA expression in chemotherapy sensitivity group after chemotherapy was 2.97±0.61, much lower than that before chemotherapy(t=4.25, p<0.05). But there is no obvious change in bcl-2 expression in chemotherapy resistance group after chemotherapy.③Cell inhibition rate and apoptosis in experimental group were 47.56%(A value:0.43±0.02)and 20.42±2.97, significantly higher than that in control group(t=3.85 and 4.80, p<0.05).④Cell inhibition rate in chemotherapy resistant group leukemia cells treated with both bcl-2-siRNA and daunorubicin was 31.06±2.30, higher than that in either daunorubicin chemotherapy group or bcl-2-siRNA group(t=4.87 and 3.64, p<0.05).Conclusions:①Bcl-2 may play an important role in the occurrence, development of B-ALL in children.②Bcl-2 may closely related with children B-ALL chemotherapy resistance, and high expression of bcl-2 may lead to leukemia cell resistance to chemotherapy, which may be one of the important reasons influencing therapeutic effects of drugs.③Bcl-2-siRNA can inhibit children B-ALL leukemia cell proliferation and promote apoptosis.④Bcl-2-siRNA can reverse chemotherapy resistance induced by bcl-2 expression. Bcl-2-siRNA has broad application prospects.
