Objective To investigate the inhibitory effects of RNAi(RNA interference) on the VEGF mRNA and protoin expression,the proliferation of Ishikawa cell lines of endometrial carcinoma,and the application of RNAi mediated by VEGF-siRNA on endometrial carcinoma.Methods On the basis of the principle of target sequence of siRNA,interference sequences of VEGF(ABO21221,GenBank) were designed.The VEGF sequence specific RNA(siRNA) and negative control siRNA were transfectd into endometrial carcinoma cell lines,Ishikawa Cell in vitro by siRNA-lipospme complex.At the 12h/24h/48h/72h/7d after transfection,RNA gene was detected by real-time fluorescence quantitative PCR(FQ-PCR);The VEGF protein was analyzed by Western blot method.MTT method and cell count were used to observe the effects of RNAi on the bioactive behalves of cells.Results After transfected,the cells,in the experimental group,deformate,shrink, and began falling out.The mRNA levels of VEGF were downregulated effectively in 12h/24h/48h and 72h respectively after the transfection of the VEGF Sequence specific siRNAs(Q=6.44/8.75/19.23/7.34,P<0.05).The protein levels were down regulated obviously post transfection compared with the control in 12h/24h/48h and 72h(F= 19.7/79.6/203.1 /58.2 P<0.05).They were significantly different compared with the control groups.After transfected,the inhibition ratio of proliferation of ishikawa cells was obviously higher than the controls'(F=20.4 /25.1/40.6/23.4,P<0.05).There was a significant difference in the levels of cell population of experimental group and the controls during 12h-Td(F=4.23/6.86/7.12/12.1,P<0.05).Conclusions The small interference RNA target VEGF can effectively downregulate VEGF on both mRNA and protein,inhibit the growth of Ishikawa cells.It hints that VEGF may plays an important role in the development of endometrial carcinoma, which provides an experimental basis for treating human tumors with anti-angiogenesis method using RNAi.
