Objective:(1)Observ the situation of restrain of lovo, conducted by LQC extracorporeal.(2) Research the apoptosis of lovo conducted by LQC.(3) Research the change of morphology to the lovo conducted by LQC.(4) Staudy the antitumor function mechanism, research the HGF/SF-met signaling transduction pathway.Methods:Culture lovo cell in vitro, use MTT to detect the inhibitory action of lovo conducted by LQC, and draw cell growth curve. Observe the change of morphology and apoptosis by Hochest33342. Use the Transmission electro microscopy to observe the morphologic changes. Study the key sites in HGF/SF-met signaling pathway. detect the content of HGF,PTK,c-met by Western Blot and real-time PCR. Use immunofluorescent staining and laser scanning confocal microscope technology to detect the expression of HGF,PTK,c-met. Confirm the influence of HGF/SF-met signaling pathway and anti-tumor mechanism of LQC.Results:We find in the vitro tests that LQC have inhibitory action to the cell lovo. In the72h, the inhibitory action is most obvious, and the inhibition ratio of LQC high-dose is31.65%. Compared with the control group, the difference has significance meaning. Transmission electro microscopy shows that the morphology has changed. Flow cytometry shows that the LQC induce tumor cell apoptosis happened. In the48h, the inhibitory action is most obvious, and the inhibition ratio of LQC high-dose is31.65%. Compared with the control group, the difference has significance meaning. Real-time PCR and Western blot were applied to study the HGF/SF-met signaling pathway. The result shows that tumor cell apoptosis happened induced by LQC is relected to the HGF/SF-met signaling pathway. LQC can restrain the union expression of HGF and c-met, and restrain HGF/SF-met signaling pathway to induce cell apoptosis.Conclusions:We find in the vitro tests that LQC have inhibitory action to the cell lovo, and induce tumor cell apoptosis happened. The result shows that tumor cell apoptosis happened induced by LQC is relected to the HGF/SF-met signaling pathway. LQC can restrain the union expression of HGF and c-met, and restrain HGF/SF-met signaling pathway to induce cell apoptosis. It provides new ideas and theoretical basis for future clinical antitumor.
