Objectives To determination the frequency and distribution of UGT1A1*28polymorphism in68small cell lung cancer patients in China.Methods Peripheral blood samples were collected from68cases of small cell lung cancer and33normal persons. Genomic DNA was extracted from the patients leukocytes in the peripheral blood. Then the target gene fragment was amplified by PCR and the genotypes were determined by analyzing the sequence of TATA boxes in the UGT1A1gene by fluorescently labeled sequencing method. The frequency and distribution of UGT1A1*28polymorphism in small cell lung cancer and normal persons in China were analyzed.Results In68cases of small cell lung cancer:51patients (75%) with (TA)6/(TA)6genotype,17patients (25%) with (TA)6/(TA)7genotype, no patients with (TA)7/(TA)7genotype. Wide-type and non-wild-type had no statistically significant differences in age, gender, clinical stage, function status score (PS score), the CEA level, the NSE level (P>0.05). In33normal persons:25patients (75.76%) with (TA)6/(TA)6genotype,6patients (18.18%) with (TA)6/(TA)7genotype,2patients (6.06%) with (TA)7/(TA)7genotype.Conclusions1. The frequency of UGT1A1*28polymorphism in small cell lung cancer in China is:(TA)6/(TA)6genotype75%,(TA)6/(TA)7genotype25%,(TA)7/(TA)7genotype0%.2. In comparison, UGT1A1(TA)6/(TA)6genotype is susceptibility gene of small cell lung cancer, UGT1A1(TA)7/(TA)7genotype is non-susceptibility gene of small cell lung cancer in China.3. UGT1A1*28polymorphism can not be a prognosis predictor of small cell lung cancer.
