In 1981, Ruan reported a anti-glycoprotein I b (GP I b) monoclonal antibody AN5 1 ,which specially inhibited the binding of von willebrand factor(vWF) to platelets. Many different monoclonal antibodies directed against distinct antigens in platelets were produced later. There are four thrombin receptors on the surface of platelets: protease-activated receptor 1 (PAR 1) , PAR4. glycoprotein I b and glycoprotein V (GP V ).Three anti-platelet thrombin receptor monoclonal antibodies were reported in France. Japan and American, but there is no report in our country. GPâ…¡-â…¢a complex are major platelet membrance glycoproteins. In order to acquire anti-platelet thrombin receptor monoclonal antibody, we immuned Balb/c using Glanzmann's thrombasthenia (GT) platelets whose OP â…¡ b-â…¢a is little. By means of cell fusion ELISA selection and several subcloning, two monoclonal antibodies SZ-16 and SZ-17 have been produced.The SZ-16 and SZ-17 were shown to be the IgOl classes by the double immunodiffusion analysis, the human platelet antigen recognized by SZ-16 had a molecular weight of 165KD under nonreducing conditions, FCM showed the binding rates of SZ-16. SZ- 17 with platelets apparently decreased after platelets were treated by thrombin,platelet aggregation test showed SZ- 16 . SZ- 17 can inhibit low concentration thrombin-induced platelet aggregation, and there are dose-dependent inhibitory effects.SZ-16. SZ-17 can't inhibit high concentration thrombin. ADP Ristocetin and arachidonic 3 acid(AA)-induced platelet aggregation. All results showed SZ- 16, SZ-17 are anti-platelet thrombin receptor monoclonal antibodies. Further studies using SZ- 16, SZ- 17 are very important to understand platelet function relationship of thrombin and platelet thrombogenesis caused by thrombin under pathological conditions, they may also be used in examining and diagnosing hemorrhagic disease. Therefore SZ-16. SZ-17 are of higher clinical significance and their perspective are very good.
