| In the present research ,wool/fur -bearing animals were chosen to study the effects of exogenous cAMP on wool follicles in vivo. One-yea-old Angora goats, neonatal Shaan Bei cashmere goats , pregnant Tan sheep (90-110 days), Arctic foxes (70-90 days) and neonatal KM mice were treated with the cAMP compound respectively .Wool and skin samples of these animals were collceted, to analyze the effects of exogenous cAMP on the growth and development of wool-/fur-bearing animals and the growth of wool fibre as well as the quality . Hair matrix cells of rat vibrissa and human scalp follicle, dermal papilla cell of rat vibrissa ,outer root sheath cell of mice vibrissa and human fetal scalp follicle were succeedly cultured ,to explore the possible mechanism and to further understand the pathway and mechanism. Two hair matrix cell system of rat vibrissa and adult human scalp follicle ,two outer root sheath cell cultured system of C57BL16 mice vibrissa and human fetal scalp follicle were established respectively ,and the effects of exogenous cAMP on these cultured hair follicle cells were investigated. Our data were as following: Effect of the cAMP compound on the growth and development of wool/fur-bearing animals and mice ,and the effects on the development and activity of wool follicles. Exogenous cAMP stimulated the growth and development of wool /fi.xr-bearing animal and mouse. The specified dose of exogenous cAMP significently increased the follicle number! unit ;Angora goat increased 8O%(p<0.01) , Cashmere goats III increasec about 46% (P<0.01).Mouse skin was thickened ,length of hair follicle and diameter of hair follicle bulb enlarged. Exogenous cAMP promoted activity of wool follicle, the experiment results demonstrated that the given dose of exogenous cAMP increaed raw wool output of Angora rams and ewes ,79. 1% and 40.9%respectivly (P<0.01 and p<0.05).Extending length of wool fiber increased 1O.8%;cashmere output of Shaan Bei Cashmere goats increased 28.2%, cashmere net length increased l2.2%(p<0.01). Exogenous cAMP was able to raise Angora rams and ewes net body weight up to 22.4% and 28.1% respectively (p<0.05);weight of the neonatal Tan sheep increased 12% and 19.8% respectively (p<0.01); 6-7-month-old Arctic foxes net body weight increased 131.4% (P<0.001);2-monthe-old KM mice body weight increased 6?2%(P<0.05). Exogenous cAMP stimulated development of mice gonad ,spermatogenic cell and ovarian granular cell of test group, ICC displayed that PCNA staining was stonger than that of the control. Exogenous cAMP promoted development of Arctic foxes pineal gland, Absolute strength of wool fiber of Angora goat improved 13% (P<0.05);Dry/dead wool ratio of test group decreased ,the value was 71.9% (P<0.01).The volume of pelge follicle bulb of test group mice enlarged. Establishment of HMC culured system of rat vibrissa and adult human scalp follcile and ORS cultrued system of human fetal scalp and mouse vibrissa folliclees Kinetics of the celles were measured. The celles identification were performed by immunocytochemisty of alpha-SMA, vimentin, CK1,4,5,6,8,10, 13,18,19. HMC was negative to all of them, ORS was positive to K19. rVMC and hHMC were subcultured 6 and 11 passages respectively ;hFORS and mORS were propagated 5 and 8 passages respecivtly. Iv... |
