Shugan Immune Hepatic Fibrosis In Rat Cells Of Extracellular Matrix Metabolism And Immune Regulation Mechanisms

Purpose: To probe into the action mechanism of Liver-Coursing and Gallbladder-Disinhibiting Method to prevent and treat liver fibrosis through studying its effects on immue regulation of T-lymphocyte and matrix metabolism; To bring to light the main pathological mechanism of liver fibrosis being liver and gallbladder stagnation.Method: The fibrosis model in rats is duplicated by injection of pig serum to abdominal cavity. We observed the effects of Liver-Coursing and Gallbladder-Disinhibiting Method from the flowing aspects: To inquire into the effect of Liver-Coursing and Gallbladder-Disinhibiting Method on collagen metabolism in rats with liver fibrodis and possible mechanism through observing microscopic morphologic changes of the fibrotic liver, determining the changes of hyaluronic acid (HA) , procollagenIII(PCin) and laminin (LN) quantitatively by radio-immunity and observing the protein expression of transforming growth factor i(TGF-ui) ,desmin and collagen III by immunohistochemical method and observing the gene exression of matrix metalloproteinase-1 and collagen Ill's mRNA by in situ hybrydization; To probe into the action of Liver-Coursing and Gallbladder-Disinhibiting Method to regulate immune function of rats with liver fibrosis and possible mechanism by detecting the changes of T-lymphocyte subsets and the ratio of CD4+/CDg+ and assaying the changes of IL-2 and IFN-v quantitatively generated by splenic lympocyte with Enzyme-linked immunoabsordent assay (ELISA).Results: There is obvious hyperplasia of fibrous tissue,widened septum interlobular and pseudolobulation of liver structure in rats in model group; The liver structure of rats in high dosage group is normal on the whole because there are no hyperplasia of fibrous tissue and pseudolobulation , but there is some vaculated necrosis in the liver. The HA levels in blank control group , high , middle and low traditional Chinese herb's group are significantly lower than that in colchicine group, and the HA level in colchicine group is significantly lower than that in model control group. The Pdll levels in blank control group , colchicine group ,high , middle and low traditional Chinese herb's group are significantly lower than that in model control group, and levels in high traditional Chinese herb's group is significantly lower than that in colchicine group. The LN levels in blank control group , colchicine group ,high , middle and low traditional Chinese herb's group are significantly lower than that in model control group. The number of CD4+ positive cell in blank control group , colchicine group ,high , middle and low traditional Chinese herb's group are significantly higher than that in model control group, and CD4+ positive cell in blank control group, high , middle and low traditional Chinese herb's group are significantly higher than that in colchicine group. The number of CD8+ positive cell in blank control group , high , middle and low traditional Chinese herb's group are significantly higher than that in model control group, but there is no significant difference in CD8+ positive cell between colchicine group and model control group. The ratio of CD/CDg in blank control group , colchicine group ,high , middle and low traditional Chinese herb's group are significantly higher than that in model control group, and the ratio of CD47CD8+ in blank control group, high , middle and low traditional Chinese herb's group are significantly higher than that in colchicine group. IL-2 yielded by splenic cell in. blank control group , high , middle and low traditional Chinese herb's group are significantly higher than that in model control group, but there is no significant difference in IL-2 between colchicine group and model control group.
