1. Study on glucose-6-phosphatase, glucokinase activities and their gene expressions in fat-fed rats of insulin resistanceAIMS: To study the activities and gene expressions of glucose-6-phosphatase and glucokinase in fat-fed rats of insulin resistance. METHODS: The rats were divided into two groups of eight rats each randomly. One group was given standard food and the other group was given high-fat diet to make insulin resistance model for 6 weeks. Insulin sensitivity was valued by hyperinsulinemic euglycemic clamp. The activities of glucose-6-phosphatase and glucokinase were measured and the mRNA expressions of two enzyme was assessed by RT-PCR. RESULTS: The body weight, TG, FFA, plasma insulin in fat-fed group was higher than that in the control group. The activities of glucokinase in fat-fed group were lower markedly than those in the control(22.4テつア10.2)mUテつキmg-1 prot VS (53.7テつア13.4)mUテつキmg-1 prot, Pテッツケツ、0.01. Compared with the control group, fat feeding significantly decreased the expression of glucokinase gene(0.36テつア0.17 VS 0.63テつア0.15,Pテッツケツ、0.05), increased activities of glucose-6-phosphatase. CONCLUTION: The increased hepatic glucose production may result from the changes of activities and gene expressions of glucose-6-phosphatase and glucokinase in fat-fed rats.
