Objective: To investigate the distribution of the single nucleotide polymorphism in exon 9 (Pro387Leu) of the protein tyrosine phosphataselB (PTP1B) in the Han nationality population in TianJin and to assess whether P387L polymorphism of the gene is associated with varying glucose tolerances subjects.Methods: A total of 327 Han people in Tianjin were involved in the study. Oral 75 gram glucose tolerance test was done among the subjects. According to the results of the test, 133 type 2 diabetes patients (68 male, 65female, average age was 54.65±11.15 ), 91 impaired glucose tolerance patients ( 50male, 41 female, average age was 54.18±11.85) and 103 normal glucose tolerance controls (52male,51female, average age was 53.05±11.06) were enrolled. The participants were genetically unrelated. Genomic DNA was extracted from anticoagulant whole blood. The exon 9 (Pro387Leu) of the PTP1B polymorphism was determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)analysis.1. There were statistics differences in waistline,hipline,WHR,SBP,DBP,FPG,2hPG,FINS,2hINS,HbAlc,TC,TG,LDL-CamongT2DM. IGT and NGT groups, and there were not statistics differences in sex,age,BMI,HDL-C and UA. 2. There were statistics difference in HOMA-β,HOMA-IR and DI among three groups. HOMA-IR was increased gradually from group NGT→JGT→T2DM,while DI and HOMA-βwere decreased step by step.3. The prevalence of the PTP1B alleles and genotypes in each group were consistent with Hardy-Weinberg equilibrium law.4. There was not LL genotype of all the subjects, same as other area in China.5. The genotype frequency of P387L polymorphism of the PTP1B gene in T2DM was 98.5% for PP,1.5% for PL, and was99.0%,1.0%in NGT group. There no mutation in IGT group. There was no significance difference in the genotypes and allele frequency of PTP 1B gene among groups.6. Three subjects with Pro387Leu mutation (PL genotype) were found that their BMI,FINS,HOMA-IR were higher than the other subjects without Pro387Leu mutation (PP genotype) , there was statistics difference.Conclusion: The results showed that the 387(P/L) polymorphism of the PTP 1B gene was not associated with varying glucose tolerances subjects (T2DM, IGT and NGT groups), but suggesting that there is possibly a relation between Pro387Leu mutation of PTP 1B gene and obesity in China.
