Objective To investigate the antithrombotic effects of ginsenoside Rg1 and its mechanisms. Methods Charlton's method was used to evaluate the antithrombotic effect of ginsenoside Rg1 on electrically stimulated rat carotid arterial thrombosis; Born's method was used to determine the effects of ginsenoside Rg1 on platelet aggregation in vitro or in vivo; The effect of ginsenoside Rg1 on platelet-leukocyte interactions was investigated according to the methods of HambuRger and Born, respectively. Results Intravenous ginsenoside Rg1 at 25 and 50mg/kg significantly prolonged the occlusion time,25,50 mg/kg of ginsenoside Rg1 prolonged the occlusion time from NS control (19.4 + 2.7) min to (30.8 + 3.4)min,(38.1 + 3.2) min respectively ( P < 0.01 ), ginsenoside Rg1 at 50 mg/kg prolonged the occlusion time from aspirin control (25.2 + 1.9)min to (38.1 + 3.2)min ( P < 0.05 ). Ginsenoside Rg1 markedlyinhibited ADP- or platelet activating factor-induced platelet aggregation in vitro or in vivo; Ginsenoside Rg1 significantly decreased the binding of thrombin-stimulated platelets to leukocytes with the 50% of inhibitory concentration (IC50) value of 73.5 (mol/L, and it could inhibit washed platelet aggregation stimulated by N-formyl-methiongl-leucyl-phenylalanine(fMLP)-activated leukocytes (IC50 = 134.3 (mol/L). Conclusion Ginsenoside Rg1 showed potent antithrombosis due to its inhibition on platelet aggregation and suppression of leukocyte-platelet interactions.
