Objective To study the effect of grape procyanidin on platelet function and thrombosis.Methods (1) GPC on platelet function of rats : Wistar rats were devided randomly into 5 groups: normal control, high fat diet (HFD) control, low dose GPC, medium dose GPC, high dose GPC. Rats of normal control were normally fed. The others were fed by HFD. The three GPC groups were given GPC by 10, 50, 150mg/Kg through mouth respectively for 6 weeks, and then the levels of serum total cholesterol(TC) ,triglyceride(TG), SOD and MDA, plasma GMP140 and TXB2, the rate of platelet aggregation were examined .(2) GPC on thrombosis in mice: Kunming mice were devided randomly into 5 groups: normal control, thrombus control, aspirin control, low dose GPC, high dose GPC The mice of aspirin control group and two GPC groups were given respectively by 50mg/Kg aspirin and 15, 150mg/Kg GPC through mouth for 8 days. On the sixth day, carrageenan was injected by hypoderm into mice except the normal control group to produce thrombus in tails. On the ninth day, the effect of GPC on thrombus were observed and GMP140 and TXB2 were examined by RIA.Results (1) GPC on platelet function of rats : The levels of lipid, serum MDA, plasma GMP140 and TXB2, the rate of platelet aggregation of high dose GPC group were significantly lower than those of HFD control group. SOD was significantly higher.(P<0.05)(2) GPC on thrombosis and platelet function: the length of black tail, percent of thrombosis frequency, GMP140 and TXB2 levels of high dose GPC were significantly...
