Objective:To observe the preventive function of Xiaoketongbikeli for peripheral nervous lesion at the early stage on diabetes rats, and probe into the mechanism of that formula.Method:Set up the diabetes rat model evoked by alloxan. According to different blood sugar values, randomly divide the rats into normal group, model group, Xiaoketongbikeli (high dosage and low dosage) group and methylcobalamin control group, respectively administer the rats with saline and Xiaoketongbikeli of different dosages by ig (1.875g/kg, 0.625g/kg), while 300 g/kg methylcobalamin for control group, one time each day. Two months after the administration, determine the FBG, body weight, SOD and MDA in blood serum of rats hi each group, observe the changes on final product of glycosylation, CGMP and CAMP of sciatic nerve and the synapsin of rats' sciatic nerves. Conduct the pathological observation on area of myelin sheath, extramedullary fiber and the cross section of myelin sheath of sciatic nerves. And observe the changes of ultrafine form of sciatic nerve through transmission electron microscope. In the mean time, determine the MNCV, SNVC and SL, and the tail-flicking test should be undertaken for checking the sensory nerve.Result:1. Xiaoketongbikeli has the function of regulating blood sugar, enhancing SOD and decreasing MDA in blood serum of the diabetes rats.2. Xiaoketongbikeli can obviously increase the content of CGMP and CAMP in sciatic nerve and the content of sciatic nerve synapsin, meanwhile reduce the final product of glycosylation in the sciatic nerve of rats.3. Xiaoketongbikeli can effectively enlarge the area of myelin sheath, extramedullary fiber and the cross section of myelin sheath.4. Xiaoketongbikeli can remarkably improve and speedup the MNCV and SNVC ofdiabetes rats, and shorten the latent period.Conclusion:Xiaoketongbikeli has the function of regulating blood sugar, enhancing SOD and decreasing MDA in blood serum of the diabetes rats, being able to obviously increase the content of CGMP and CAMP in sciatic nerve and the content of sciatic nerve synapsin and reduce the final product of glycosylation in sciatic nerve, and effectively enlarge the area of myelin sheath, extramedullary fiber and cross section of myelin sheath of rats' sciatic nerve, and remarkably improve MNCV and SNVC of diabetes rats and shorten the latent period. It can obviously ease up the pathological changes of peripheral nerve at the early stage of the diabetes rats, and the overall curative effect is better than that of methylcobalamin.
