AIM: The present study was designed to study the effect of PPC on coronary heart disease and angina pectoris.as well as its mechanism.Modern methods of pharmacology and molecular biotechniques were used to research the target genes of PPC based on its pharmacodynamics and toxicology. METHODS: All methods of pharmacology were in accordance with the guidance of R&D of New drugs of SFDAjncluding the effect of PPC on myocardial ischemia in dogs and rats,acute toxicity and chronic toxicity.The techniques of gene chips ,as well as the other molecular biotechniques such as immunohistochemistry study, in situ hybridization,RT-PCR were used to study the mechanism of PPC .RESULTS: PPC can alleviate the status of myocardial ischemia in dogs and rats,enhance the flow of blood in coronary artery and cardiac output.reduce the release of myocardium enzyme such as CK, ATPase, SDH, lack the blood cardiac muscle to consume oxygen ,and it can also reduce the conglutination of the blood platelet, suppress the formation of thrombus .In this study,47 different genes of which expressed between normal group and model group were selected,as same as 205 different genes between the treated group and model group, 102 different genes between treated group and normal group,27 genes of which were studied in this project were confirmed to have relations with the pharmacodynamics of PPC.At the same time.the selected target genes such as Hsp70, eNOS, HO-1 and HO-2 were studied by RT-PCR, in situ hybridization and immunohistochemistrystudy.CONCLUSION: Myocardial ischemia in dogs and rats could be alleviated by PPC. They have also shown protective effect on cardiomyocytes of neonatal rats during hypoxia.lt was a kind of safe drug tested by acute toxicology and chronic toxicology.The part of its target genes were Hsp7, eNOS, HO-1 and HO-2.
