Tissue repair is the hotspot of surgical researches at present. In clinic repair obstruction mainly showdelayed healing wound (DHW), such as diabetes mellitus accompanied skin ulcer, venous leg ulcer andpressure skin ulcer, et al. Inflammation is the vital phase of wound repair, without it without healing.Based on long-termeffective prescription of Beijing Chinese medicine hospital, YangYuTuJi (YYTJ) wascomposed of Astragalus Polysaccharin(APS) , Perlolyrine, Cinnamylaldehyde and Muscone under theprinciple of supplementing Qi ,warming Yang, promoting blood circulation and accelerating capillarymovement. The mice with immunosuppressive and skin-wound (ISW) were adopt to investigate themodulating effect of external usedYYTJ on wound healing.1Experiment in vivo1.1 Duplication of the wound with immuno-suppressive mice induced by systemic hydrocortisone— Characteristic withYang deficiency inTCM Balb/c mice were attacked by systemic hydrocortisone for 7 days, and then a diameter 11mmfull-thick skin was excised on the back of mice. General condition, wound area, and histologicalcharacters were observed, as well as the proliferation of T and B lymphocyte by MTT assay,phagocytosis of Mφ in abdominal cavity by neural red assay were investigated respectively. The resultsdemonstrated: the mice attacked by systemic hydrocortisone showed a depressed immune condition andimpaired wound healing. The proliferation of Tand B lymphocyte and phagocytosis of Mφ in abdominalcavity were significantly inhibited, as well as the number of inflammatory cells and new collagendeposition was less than control group. Wound area was larger than control group at post-injury day 7.The results indicate that the wound of skin excision complicated with hydrocortisone attack were still inimmunosuppression and delayed healing condition. In characteristic they are accordance with Yangdeficiency inTCM..1.2 The effects ofYYTJ on wound healing of mice with I SWand the mechanismstudies. YYTJ of high dose(1.05g/L),mediumdose(0.53g /L)andlowdose(0.27g/L)was appliedon the mice with ISW. Wound area and healing rate were analysed by Image-proplus sofeware,histological characters described by HE and Masson, expression of TGF-β1 and CD68 stained byimmunohistology assay, as well as IL-1β, IL-6, IL-8 and TNF-α levels measured by radioimmunoassayrespectively. The results demonstrated: (1)YYTJ reduced wound area,increasedhealingrate.(2)YYTJincreased the number of Mφ in wound area(3) YYTJ regulated the levels of IL -1β,IL-6,IL-8 andTNF-α levels in injury tissue. (4) YYTJ increased TGF-β1expression and new collagen deposition ininjury tissue. The results indicate that YYTJ can promote wound healing by regulating inflammatoryresponse in the wound of mice with ISW.英文摘要 V2 Experiment in vitro2.1 Effects of components of some Chinese herbs on proliferation of human umbilical vein endothelial cells (HUVEC) in vitro To investigate the effects of some components of external use herbs on proliferation of HUVEC.MTT assay was used to observe the effect of Rg1, Rh1, Perlolyrine, Cinnamylaldehyde, Muscone,AstragalusPolysaccharin, Velver antler polypeptide(VAP) and soluble extract of Boswellia carteriiBirdw(BCB) on proliferation of HUVEC. The results demonstrated: APS did not show the promotingeffect on HUVEC proliferation at the concentration of 9.75mg/l-2.5g/l. Rh1 promoted HUVECproliferation at 1.94mg/l-0.5g/l(p<0.05 , p<0.01 respectively).Rg1 inhibited HUVEC proliferation at31mg/l( p<0.05). VAP promoted HUVEC proliferation at 1mg/l-0.5g/l with optimal dose of 10mg/l(p<0.01).Cinnamyl aldehyde promoted HUVEC proliferation of at 2g/l(p<0.05).Muscone andsoluble extract of BCB inhibited HUVEC proliferation at 1g/l, 0.5-2.5kg/l (p<0.01 respectively).PerlolyrineinhibitedHUVECproliferation at 0.125g/l-0.5g/l(p<0.01 respectively).Theresultsindicate:The external herbs for supp...
