Objective: To discuss the effect mechanism of mitocondria on apoptotic signal transductional pathway, through investigating mitochondrial shape, structure, regulation among membrance protein and mitochondrial DNA deletion. Meantime, to investigate the effect on prolong brain aging by moxibustion the points Baihui(DU20), Shenshu(BL23).Methods: On the base of nature aging rats model, the rats were divided into 3 groups: adults group, aging group, and moxibusition aging group, the latter were treated by moxibustion Baihui(DU20), Shenshu(BL23) points in 10min, one a day , 5days a course of treatment, all 8 courses. Using several measures including electro-glasses, flow cytometry(FCM), immunohistochemical method, polymerase chain reaction, and Western blotting, et al, to observe each group's alteration on hippocampi nervous cell and mitochondrial ultrastructure, the mitochondrial membrance electro-bit and the ratio of nervous cell apoptosis. And to test some signs' change on Bcl-2/Bax, Caspase-3,-9, mitochondrial DNA,14-3-3 protein.Results: (1)Through moxibustion 8courses on Baihui(DU20), Shenshu(BL23), the moxi-aging group has no difference(P>0.05), compare to the adults group, on average volume, body-density, quantity-density, superficial ratio and crest membrance density. But compare to aging group , the moxi-aging group has distinctive difference(P<0.01), which shows the moxibustion could restain mitochondrial figure-structure changes , maintain its function, and prolong brain aging. ( 2 ) The moxi-aging rats were prevented the dissipation on hippocampous A Ψmt and reduced the number of apoptosis by moxibustion treament, which has clearly difference (P<0.01) comparing the aging group; The correlation analyse indicates that there exsists a linearity negativecorrelation between apoptosis and △Ψ mt ( r= -0.483, P<0.01), which affirms the relation on mitochondria and cell apoptosis in the process of senescence. ( 3 ) Through moxibustion , the Bcl-2 protein occurs strong masculine expression on the moxi-aging group, compared to aging group and adults group ,which has obvious difference(P<0.01); and Bax protein happens weak expression, which causes the difference to aging group is P<0.01, to adults group is P<0.05. but at last compared to the aging and adults groups ,the obvious difference on the ratio of Bcl-2/Bax is P<0.01. ( 4 ) Compared to the aging group and adults group, the moxi-aging group could differently reduce the activity of caspase-3 and caspase-9(P<0.01). meantime, the expression on caspase-3 protein is reduced bymoxibustion . ( 5 ) The mtDNA deletion on nervous cell of aging rat is clearly higher than the adults and moxi-aging groups, which shows mtDNA is firmly relate to age increasing. But through moxibustion, the mtDNA deletion is lessened, the difference among the moxi-aging group, adults group and aging group separately is P<0.05 or P<0.01, which shows moxibustion "Baihui", "Shenshu" has protection for mtDNA deletion. (6) 14-3-3 protein declines with age increasing, because the content of 14-3-3 protein to aging group is lower than to adults group(P<0.01); but the phenomenon could be improved by moxibustion that is the content of 14-3-3 protein to moxi-aging group is better than to aging group(P<0.01), which clews the moxibustion treatment could restrain 14-3-3protein's change with age increasing and enhance its protective action.Conclusion: From the whole point of view that mitochiondria-apoptosis-aging, mitochondrial change on figure and structure is a switch implement of apoptosis and a center tache to senescence. While moxibustion on Baihui and Shenshu may not only significantly improve and protect mitochondrial structure and function, but also effectively adjust the signal transduction system with polylink and polytarget of apoptosis with the center of mitochondria, so as to control the apoptic progress and prolong the brain aging.
