Depression is a mood disorder or emotional disorder. The cause of depression, although still not completely understood, stems from multiple sources. Clinical depression is a psychological condition that changes how you feel and think as well as behave. Due to its complexity, the pathology of depression remains unclear in spite of many studies performed in the subject area. In recent years, many types of anti-depression medications came to the market; however, their effectiveness is limited to controlling certain symptoms. Other than the unwanted side-effects such as increased toxin in blood stream, the patients often exhibit behavioral problems such as resistance to following orders. Acupuncture has been used to treat depression for some time and proven to be effective to certain degree. Unlike medication therapy, acupuncture treatment for depression has no known side-effect. Moreover, it proves to promote modulation of, and coordination between physical components. Some of the acupuncture clinical trials have shown promising results in treating depression.Recently, the relationship between the high levels of mental activity and the nerve-endocrine-immunity modulation system has attracted the interest of many researchers and become one of the hottest research topics in the past few years. It is known that one's mood can affect his or her social behavior and sense of physical well-being as well as mental health. Unwelcome life events, especially stressful ones, can trigger a depressive episode. The Hypothalamic-Pituitary-Adrenal axis (HPA axis) and the Autonomic Nervous System (ANS) are the two most crucial parts of the brain in response to stress. The neuroendocrine hypothesis of depression is that the depression patients had notable HPA hyperactivities. Based on this theory our approach focuses on how electroacupuncture (EA) in conjunction with traditional acupuncture therapy will effect the HPA axis so as to determine the effectiveness of the treatment.We make the grade standard based on the principal rule of clinic research in DME. From the literatures, we try to find the frequent most of the acupoints. We choose the long-time unpredicted chronic stress model that can simulate the predisposing cause of the depression circumstance and stress hormones changes as the research object. To analysis the different kinds of hormones in three tissues by RIA method. To detect the relative genes expression in the cell signal transduction route and relative receptor expression in the negative feedback of HPA axis by real-time quantitative reverse transcription-polymerase chain reaction method. Through the quantity changes of HPA axis hormones and the pathological changes of the negative feedback in the stress rats by the methods above, we suggest to discuss the mechanism that effects of E A on the HPA axis.1. Based on the RCT principle of DME, we found the frequency of Bai Hui and Yin Tang points used is highest in the EA therapy.2. The chronic stress rats are in the depressing states. The chronic stress can cause the HPAaxis functional hyperactivity and the disorder of the negative feedback. The symptoms and pathological changes of the binding group rats which are given the binding stress in the same extent with the EA group rats are more servious than that of the model group rats.3. Fluovetine can alleviate the depressing state of the stress rats and regulate the hyperactivity of HPA axis.4. The EA therapy can alleviate the state of the depressing of the stress rats obviously. It can reverse the vicious circle of HPA axis by several joints. It can affect the cytokine, hormone and hormone receptor. We speculate the effect of EA on the nerve, endocrine, immune system is to make them comformed. This is the advantage of the EA compared with the west medicine. At the same time, we conside that there are more effect points of EA therapy than taking the Fluovetine.5.
