Objective: To find out the differential expression genes in gallbladder carcinoma by profiling cDNA microarray and to provide data for the diagnostic cDNA microarray. The validation of the microarray was confirmed simultaneously.Methods: The fresh specimens of gallbladder and normal control were preserved inCryopreservation, then the total RNA was extracted and detected. Hybridization with BiostarH-40s type profiling cDNA microarray was performed after the RNA was labeled with fluorescent probe. The image scanning and data analysis were performed subsequently. Finally, the results of hybridization were validated by real-time PCR.Results: 61 differential expression genes were definited in all 4 specimens, which wasinvolved in several aspect of tumor development. There were 17 genes down-ragulated and 44 genes up-ragulated. There were 9 oncogene and anti-oncogene; 7 genes about apoptosis; 6 genes about Synthesis, repair and reconstruction of DNA; 13 genes about signal transformation; 12 genes about metabolism and others 8 gene. 5 genes among them were selected to be amplificated, and the expression of them was coincident with the microarray.Conclusions: Gene expression profiling in human gallbladder carcinoma can beanalyzed by cDNA microarray sensitively, generously, and effectively. The result of the experiment of microarray was reliable.
