Uterine Leiomyomas is the most common smooth cell tumor of the myometrium. This thesis intends to apply the method of "promoting blood circulation, lubricate yin and clear heat as to dissipate blood stasis". It based on my professor say the disease is "chong and Ren channels stasis ,and transforming into heat and yin is vacuous, concretions occur as a result" of the TCM pathgensesis in clinical experience.Our research is using the serum containis different concentrations of Chinese medicine " Gonhliuqing capsule"(7.2g/Kg,3.6g/kg,1.8g/kg) in comparision with mifepristone,ru486. (2.5mg//kg) add on the human uterine leiomyoma cells in vitro. Isolated human leiomyomas cells were subcultured in DMEM supplemented with 10% FBS . MTT assay for cell growth and viability(10h~40h,24h~144h). Immunocytochemical staining for ER, PR, the growth factor receptors IGF-1R, EFGR expresstion (20h,48h,72h,3 times medical serum added). Flow cytometry detect the Gonhliuqing effects on the cell cycles and apoptosis of the leiomyomas cells (20h, 48h, 72h, 3times medical serum added).The result shows that: 1. Gonhliuqing suppressing on proliferation of uterine leiomyomas cells, in the 3.6g/kg status had the most significant effect. 2. Gonhliuqing had the effect on regulating the ER, EGFR and IGF-1R, but not on PR. 3. Gonhliuqing tunning the leiomyomas cell cycle and induced apoptosis. In summary , we report that Gonhliuqing can prevent the formation and development of leiomyomas , its mechanism concern with regulation ER, IGFR and EGFR, district cell cycle and induced apoptosis of leiomyomas cell.
