Objective Insulin-like growth factor-II (IGF-II) is potent mitogen and postulated to exert autocrine/paracrine effects on growth regulation in human cells. It is an imperative stimulating factor for malignant cell growth from G1 to S stage. After specific binding with its receptors(insulin-like growth factor- I receptor/insulin-like growth factor- II receptor, IGF- I R/IGF- IIR), IGF- II plays an important role in promoting proliferation, differentiation of malignant cell through a series of signal transmission, and participates occurrence, development, metastasis and new vessels formation of tumor accordingly. In order to establish experimental and theoretical foundation for gene therapy of GC such as anti-IGF- II Mab, anti-IGF- I R Mab, IGF- II antagonist and antisense RNA target therapy, we evaluated the levels of serum IGF- II, and the variation perioperatively of serum IGF- II and the expression of IGF- II, IGF-I R, IGF- II R in patients with gastric carcinoma (GC), investigated the relationship between the expression of IGF- II, IGF- I R, IGF- II R and the occurrence, development, histological type, infiltration, metastasis, TNM stage and helicobacter pylori (Hp) infection of GC, Observed the influence of anti-IGF- II monoclonal antibody (Mab) on morphologic and characteristic ultrastructural and cell cycle and apoptosis of BGC-823.
