Glycyrrhizin level is one of the most important indexes to estimate the quality of medicinal licorice. However, it varies among different habitats, individuals , havest times and parts. These differences make the clinic effect of medicinal licorice instable, so it is significant to reveal the mechanism of the variation of glycyrrhizin content.In order to elucidate the mechanism of the formation of glycyrrhizin, the paper compared the differences of ITS sequences andβ-amyrin synthase gene (No.1 extron) between the glycyrrhizin group and no-glycyrrhizin group. 0n the basis, the differences of ITS sequences andβ-amyrin synthase gene (No.1 extron) among the populations are compared to interpret the mechanism of the accumulation of glycyrrhizin. Furthermore, the expresses of theβ-amyrin synthase gene are compared among the different parts of glycyrrhiza uralensis.The important conclusions are as follows:(1) Based on the differences of ITS sequences, the two groups of the glycyrrhiza ssp. , the glycyrrhizin group and the no-glycyrrhizin group, are TTCCTG genotype and ACTTGT genotype,respectively,And according to 5.8S sequences ,the glycyrrhizin group and the no-glycyrrhizin group can be A and G genotype,respectively. As what we predicted , the genotype divided on the basis of ITS sequences and 5.8S sequences, are highly correlated with the formation of glycyrrhizin.(2) The ITS sequences of Glycyrrhiza inflata and G.glabra are the same as that of G. eurycarpa and G. glabra L. var. glandulosa respectively,which strongly supports the classification treatment of the combining of Glycyrrhiza inflata and G. eurycarpa, G.glabra and G. glabra L. var. glandulosa. The result shows that the G. eurycarpa and G. glabra L. var. glandulosa can be used as the resources of medicinal licorice.(3)β-amyrin synthase gene exists in all of the glycyrrhiza plants, either glycyrrhizin group or no-glycyrrhizin group,which proves the fact that there is hardly any correlation between the absence ofβ-amyrin synthase gene and the unformation of glycyrrhizin.Inversely the variation ofβ-amyrin synthase gene is critical for the biosynthase of glycyrrizin.(4) There are rich variation ofβ-amyrin synthase (No. extron) among the different population of glycyrrhiza uralensis, six genotypes are discovered in 19 accessaries, which are type I(TATGAGGT),type II(TATGGGGT),type III...
