Part One. Distribution and expression of Peroxisome Proliferator Activated Receptor y in Pituitary adenomas and cell lineObjective To evaluate the distribution and expression of Peroxisome Proliferator Activated Receptor γ (PPARγ) in pituitary cell line and adenomas from 38 consecutive pituitary adenomas patients and 5 normal pituitary tissues.Methods Thirty eight consecutive surgically resected pituitary adenomas and 5 normal pituitary tissues were enrolled in the study. Immunohistochemistry was used to confirm the distribution of PPARγ. Expression of PPARγ was evaluated by Western Blot. PPARγ expression of GH3, MMQ and ATt20 cell line was evaluated by RT-PCR and Western Blot.Results PPARγ immunoreactivity was located in nucleoli of pituitary adenoma cells. PPARγ was expressed in all human pituitary adenomas and normal pituitary tissues. The expression in pituitary adenomas was significantly higher than in normal pituitary tissues (P<0.01). There was significant difference of PPARγ expression between invasive and non-invasive adenomas. There was high expression of PPARγ in GH3, MMQ and ATt20 cell line.Conclusions PPARγ may play an important role in the growth and invasion of human pituitary adenomas and maybe a novel therapeutic target of pituitary adenomas.
