The Active Substances Of Traditional Chinese Medicine Radix Screening Is Mainly Based On The Expression Of Biological Thermodynamics And Quality Evaluation

In this paper we try to explore a new mode of active substance screening and quality evaluation under the guidance of TCM theory. Thermodynamics was used as main technique to emphasize the metabolized process and index of mutual reaction between radix isatidis and the organism. Ultimately a novel method and mode of active substance screening and quality evaluation mainly based on the theory of biothermodynamics was brought forward and tried to establish. Results as following:1. Chemical fingerprint chromatographs of hetero-polarity extractions of Radix isatidis were established. Sample materials came from different locations, such as Anguo Hebei province, Ruobu Jiangsu province, Nantong Jiangsu province and Lingquan Anhui province. After systematical investigation, the HPLC fingerprint of petroleum ether extraction, n-butanol extraction, ethyl acetate extraction and residual in Radix isatidis were established.2. The chemical characteristics of the total organic acid part and total polysaccharides part inRadix isatidis were analyzed and the HPLC fingerprint of the total organic acid part was established. Use principal component analysis as main statistics, the internal integral area as the samples, statistical results indicated that the difference of the HPLC fingerprint mainly related to the content of total organic acid, peak 3~# (benzoic acid ) and peak 5~# (salicylic acid).3. Polysaccharides in Radix Isatidis were fractionated by DEAE cellulose ion exchange chromatography and SephadaxG 75 gel chromatography. HPGPC, IR, TLC and UV were used to investigate the structural and physico-chemical property of the homogeneous polysaccharide parts. Finally compounding basis was confirmed.4. Adopting the method of biothermodynamics, biothermodynamical chromatographs of the aqueous extracts, hetero-polarity extractions, the total organic acid parts, the total polysaccharides parts and the compound of different chemical parts in Radix isatidis was established.5.
