The patho-factor of pulmonary fibrosis(PF) is very complex. With the development of cytobiology and molecular biology there are much proof shows PF is a disease induced by multiple factor, multiple organ and multiple cell; and happens between cells and mesenchyma in multiple path. The elucidation of pathogenesis of PF is helpful to pharmacal research, especially drug screening. The drugs that is being used in clinical treating PF,eg: hormones and immunodepressant, are short of curative effect and have severe adverse re reaction. The abstraction and separation technique of active component in Chinese matera cedica have developed rapidly. To search for active component or monomor of definite pharmaco-action is the trend in field of healing PF with tradition medicine.The central content of my topic is to screening active component or monomor of definite pharmaco-action of inhibiting PF according to traditional medicine, and observing the intervention to rat model of PF induced by bleomycetin. We have established experimental foundation for researching new type of drug to healing PF.1. First, we screened the seven kinds of crude extract (paeoniae radix, ilexpubesceus, plumula nelumbinis, euphorbiae fischerianae, maoyancao, euphorbiae and taraxaci), by primarily culturing rat lung fibroblast. We have found that plumula nelumbinis, euphorbiae fischerianae and euphorbiae can inhibit the proliferation of rat lung fibroblast. Plumula nelumbinis is the best drug with most effective and little cytotoxic action. Then we screened the different extract part of Plumula nelumbinis. At last the result of screening showed that the most effective component was phenol totalbase of plumula nelumbinis.2. Secondly, we established the method of HPLC to determine the content of liensinine in order to control the quality of phenol total base of plumula nelumbinis. With the method we compared three different solvent extraction, and the result showed alcohol-acid water-organic solvent was the best solvent extraction. Extraction percentage of total base is 11.25%, and extraction percentage of liensinine was 1.20%. The solvent extraction is simple handing.3. Finally, we established rat model of PF induced by bleomycetin, and observed the interventional action of phenol total base to model. The analysis of experimental result including pathematologic and tissue biochemical indicator showed phenol total base of plumula nelumbinis have some therapeutic action to rat model of PF induced by BLM. The mechanism of action maybe include the depression in content of collagen, inhibition to proliferation of lung fibroblast, action of anti free redical and anti lipid peroxidation, and elevation of content of GSH-PX.So we think phenol total base of plumula nelumbinis is a sort of promising drug to healing PF. But its detailed and complete of mechanism of action need more relative reseach..
