Background: It has always been an important issue to protect ischemic myocardial cells in cardiology. Some Chinese herbs are helpful to myocardial ischemic diseases because of their lower side-effect and multi-targeted effect. Liposome is an ideal carrier for medicine. It will be hopeful to get a better effect when liposome is used to trap some Chinese herbs which have an effect on ischemic myocardial cells.Objective: To prepare allitridum long circulating liposome, observe its effect on ischemic myocardial cells, and study the mechanisms on which it works. Methods:1 Pharmaceutical preparationâ‘ Preparation of liposome: Allitridum long circulating liposome(ALL) and allitridum liposome(AL) were prepared by a reverse-phase evaperation technique. The entrapment ratio was detected by acentric method, the morphology was observed by transmission electric-microscope, the size distribution was detected by dynamic laser dispersion method, and the stability of the liposomes was observed for 3 months.â‘¡Irritation of the drug: 9 rabbits were randomly divided into 3 groups(allitridum injection group, AL group, and ALL group). Drugs were injected into the vein of left ear and the muscle of left leg, or dropped into the left eye of rabbits respectively, while saline solution was given to the right side of corresponding places as comparison. The vessels, muscles and eyes were observed 10, 30, 60, 120min, 24h, and 48h after the last time when drugs were given, and the tissues were collected for further observation by microscope.â‘¢Detection of drug concentration in serum and heart tissue of myocardial ischemic reperfusion injury rats: 18 SD rats were randomly divided into 3 groups (allitridum injection group, AL group, and ALL group). Rats were subjected to 30 minutes of ischemia followed by 3 hours of reperfusion by ligating or releasing anterior descending branch of coronary artery. Drugs were injected intravenously 15 minutes before reperfusion, and the sample of blood and myocardium were gathered 3 hours after reperfusion.
