The acute lower limb ischemia is one of the challenges for blood vessel surgeons. Its incidence rate approximately is 1.7 cases /10000 person per year. Although the diagnosis and the treatment aspect of the disease has obtained the very big improvement in recent years, its mortality rate and the amputation rate are still quite high. Two-dimensional electrophoresis technology is the classical method to study proteomics at present. With the widespread application of mass spectrometry technology to the study of biological macro-molecule research, proteomics study is becoming the hot spot in research. In this research, two-dimensional electrophoresis, the computational image analysis and mass spectrometry(MS) are used to study protein expression in blood vessel from acute lower limb ischemia, which will be helpful to to promulgate the pathogenesis for the disease and will develop the new mentality for better diagnosis and the treatment.First, acute lower limb ischemia animal model are established on rabbit, obtain anminal's ischemic blood vessel in different ischemic time. 2-D electrophoresis is performed after protein extraction and quantification. Obtain blood vessels gelatin atlas for rabbits' normal and ischemic blood vessels after 4 hours, 8 hours, 12 hours and 24 hours by the technique of silver stain. After computational image analysis and allocated proportion, results are: the number of total protein bands are 225 ±36, 256 ±45, 254±41, 269±52 in different ischemic time respectively, average matching rate is 64.4%; average deviation is 0.0498± 0.0268 for rabbits' normal and ischemic blood vessels after 4 hours, 8 hours, 12 hours and 24 hours, and average deviation of molecular weight: 0.9655 ± 0.2230. Qualitative and quantitative analysis for the atlas is a well-accepted method in the study of 2-D electrophoresis.
