1998 saw the publication of two papers describing in vitro the growth of human embryonic stem cells derived either from the inner cell mass of the early blastocyst or the primitive gonad regions of early aborted fetuses. People realized the worth of embryonic stem cells in the regenerative medicine. The realization of such pluripotentialty resulted in the genuine belief that stem cells research will drive a revolution in terms of how we treat many kinds of diseases. With the development of science, the adult stem cells, which bear characters of self-renewed, proliferation and multiple of differentiation potential, were isolated from fetal and postnatal tissues and organs. Resent studies demonstrated that adult stem cells may be able to repair the injured tissues. The great breakthrough of adult stem cells was that they can differentiate into other tissues' cells, not only cells of other lineges but also cells of other germ layers. This characteristic of adult stem cells was called as stem cells plasticity.The plasticity of adult stem cells attracts many researchers, especially the study of mesenchymal stem cells (MSCs) derived from bone marrow(BM).MSCs could differentiate into various cells of three germ layers. Meanwhile the character of MSCs in inducing immunotolerance provided the possibility of utilizing MSCs in tissue engineering. These cells hold tremendous promise as model systems for development of new therapeutics and development of replacement tissues once we through understand their differentiation mechanism. On the other hand, if MSCs reside in diverse tissues and organs? What other roles do MSCs play? Do MSCs have some relationshipe with certain disease processes? And how to regulate the differentiation of MSCs? All these questions are our purposes to investigate in this paper.Initially, several monoclonal antibodies were used in the isolation and characterization of MSCs, including SH2, SH3 and SH4.
