BackgroundCardiac hypertrophy is one of the most common complications of hypertension, and it is an independent risk factor of cardiovascular disease. Cardiac hypertrophy includes cardiomyocytes hypertrophy, cardiac fibroblasts proliferation, extracellular matrix production and accumulation. Apart from mechanic stimulation of overload, the role of neurohumoral factor and vasoactive peptide in cardiac hypertrophy is inspiring. It is found that cardiac cells secrete vasoactive peptides such as angiotensin â…¡,endothelin-1, atrial natriuretic peptide and adrenomedullin by an autocrine or paracrine fashion, which has an important role in regulating cardiocytes hypertrophy or proliferation in local situation. Vasoactive peptide not only have an effect on cardiocytes by itself, but also have an interrelationship in their synthesis, secretion and function between themselves, which makes cardiac remolding regulated and controlled by several substance through net work. Although it was studied in depth about one vasoactive substance on one type cardiocytes, limited information on their interrelationship in regulating cardiac hypertrophy has been had until now. There are many questions needed to be clarified. It is significant of this work for deeply knowing the mechanism of cardiac hypertrophy and the development of new treatment. So, present study is designed to investigate the most important vasoactivepeptide as Ang, ET-1 and ANP--their regulating effect on cardiocytes and themechanism. ObjectIn cultured cardiomyocytes and cardiac fibroblasts of neonatal rat, it is designed to investigate the effect of Ang, ET-1 and ANP on cardiomyocytes hypertrophy and cardiac fibroblasts proliferation single or combined, and the interrelationship of their synthesis.
