[Objective] To detect the expression of PRL-3 gene in the primary lesion and regional lymph nodes, analyze the patterns of gene expression and evaluate its clinical significance. [Materials and Methods] The specimens of colorectal cancer were continually collected from Apr 2005 to Jul 2005, and some specimens of gastric cancer were also collected as control. Real time PCR technique was applied to test the gene expression of PRL-3 quantitatively. SPSS 13. 0 software package was used in statistic analysis. [Results] Totally 192 samples were collected, with 47 primary colorectal cancers, 28 metastatic lymph nodes and 63 non-metastatic lymph nodes; 17 primary gastric cancers, 17 metastatic lymph nodes and 17non-metastatic lymph nodes. PRL-3 gene expression in metastatic colorectal lymph nodes was higher than that in the primary and non-metastatic lymph nodes. But similar result was not found in the gastric cancers. [Conclusion] Expression of PRL-3 shows remarkable specificity of tissue and correlation with metastasis.[Objective] To analyze the correlation between expression of PRL-3 gene and clinicopathological characteristics in the primary colorectal cancer. And to evaluate the prognostic significance of PRL-3 gene expression in the patients with colorectal cancer. [Materials and Methods] The information of 196 patients who received operation from Jan 1998 to Dec 2000 was collected and the expression of PRL-3 gene was tested by means of immunohistochemical technique. Statistic analysis was fulfilled by the SPSS 13. 0 software package to analyze the correlation and prognostic value. [Results] The high expression of PRL-3 happened in 97 of 196 cases (49. 5%), and it was closely correlated with operation method, differentiation, vascular invasion. Dukes' stage and metastasis. And uni-variate and multivariate analysis approved its prognostic significance. [Conclusion] Expression of PRL-3 gene is a new and significant prognostic factor of patients with colorectal cancer. High expression of this gene suggests the possibility of postoperative metastasis and shortening of survival.
