Objective:To:analyse genetic pattern and genetic heterogeneity and to investigate the relationship between Calpain10 gene UCSNP43 and UCSNP44 polymorphism and diabetes in familial type 2 diabetes mellitus of Uygur in Xinjiang.To investigate and describe the dietay pattern and nutritional condition of Uygur residents of Kashi city in Xinjiang.Methods:We chose 96 Uygur type 2 diabetic families and apply PPAP statistic software;We chose 62 Uygur type 2 diabetic families and apply PPAP and SPSS13.0 statistic software to analyse the genetic.heterogeneity about age,waistline,body mass index,blood pressure,plasm lipid.619 subjects from 75 Uygur type 2 diabetic families were divided into groups including control group and dianbetes group and were analysed in familial members.The polymorphism of Calpain10 gene UCSNP43 and UCSNP44 measured by SNP-Stream of gene chip method.SPSS13.0 statitic analysis software was used to test the relationship between Calpain10 gene UCSNP43 and UCSNP44 polymorphism and diabetes in familial type 2 diabetes mellitus of Uygur in Xinjiang.422 residents were investigated and their individual intakes were obtained by using weighing method and 24-hour recalls method..Results:1)The heritability of the first degree relative in Uygur familial type 2 diabetes was 0.578 and of the second degree relative was 0.032.With the down of the degree relative,the heritability was decreased.2)Analysis of Penrose's indicated observed value was approximate to 1/q1/2=2.7(s/q =2.44 in men,s/q =2.35 in women),so we accepted MF the genetic pattern of Uygur familial type 2 diabetes was multifactorial inheritance.3)There had significang different in body mass index,blood pressure,total cholesterol and triglyceride among type 2 diabetic families. Inaddition to age of onset,waistline,body mass index,blood pressure,plasm lipid had genetic heterogeneity between proband and relatives.4)The frequencies of UCSNP43 and UCSNP44 genotype and allel were tested by Hardy-Weinberg equilibrium(χ2=1.91, P=0.17 andχ2=1.14,P=0.70).5)There was significant difference(χ2=10.42,P=0.007) among the three genotypes of UCSNP43 in two groups.The carriers of AA genotype had higher fasting insulin and BMI than GG and AA genotypes carriers.There was no significant difference(χ2 =3.67,P=0.16)among the three genotypes of UCSNP44 in two groups.The distribution of UCSNP43 andUCSNP44 allelic frequencies had no significant difference between two groups.6)We divided all subjects by gender.The carriers of AA genotype had higher the fasting insulin and HOMA-IR than GG and AA genotypes carriers in men(F=8.89 P=0.00 and F=4.92 P=0.008).The carriers of AA genotype had higher BMI than GG and AA genotypes carriers in women(F=4.7 P=0.01).There was no significant difference(χ2 =3.67,P=0.16)among the three genotypes of UCSNP44 in men. The carders of CC genotype had higher TG than TT and TC genotypes carriers in women (F=6.46 P=0.002).7)We divided all subjects by BMI,abdomen circumference and having or not having metabolic syndrome.There were no significant difference among the three genotypes of UCSNP43 and UCSNP44.8)The citywide average of energy intake was 2381.46kcal per man per day,of protein was 68.95g,and fat was 49.07g. The average consumption of rnicronutrtion were low,especially of vitamin A and calcuim. The proportion of all kinds of fatty acid was unreasonable.Conclusion:the genetic pattern of Uygur familial,type 2 diabetes was multifactorial inheritance.With the down of the degree relative,the heritability was decreased;Body mass index,blood pressure,total cholesterol and triglyceride probably had genetic heterogeneity among Uygur type 2 diabetic families in Xinjiang.The Calpainl0 gene UCSNP43 polymorphism might be associated with diabetes in Uygur.The carriers of UCSNP43 AA genotype might have much sever insulin resistence than GG and AG genotype.The Calpain10 gene UCSNP44 polymorphism might not be associated with diabetes in Uygur.The irrationality of proportion of micranutrition and absence of micronutrition are resulted from specific dietary pattern of Uygur people.It is essential to adjust or change it.
