Objectives This study was to investigate the distribution of the gene polymorphism in CAPN 10 UC SNP43 G/A in the Han nationality population in Tianjin,and to assess the relation between CAPN10 UC SNP43 G/A polymorphism of the gene and type 2 diabetes mellitus(T2DM) and insulin resistance.Methods A total of 380 Han people was involved in the study.The participants included of 125 control subjects( males,68 females,57, average age was 55.57 + 10.93), 255 T2DM patients(males,128,females,127,average age 58.77 + 8.85 ).T2DM patients were recruited among in-patients in our hospital during the period of time May 2002 and October 2004.People who underwent a check-up examination in our hospital during the period of time between October 2003 and January 2004 were enrolled as random population control. The participants were genetically unrelated and were all living in Tianjin .Genomic DNA was extracted from the collected blood samples,using Puregero DNA isolation kit .the UC SNP43 G/A polymorphism was determined by polymerase chain reaction-restrition fragment length polymorphism(PCR-RFLP) analysis.The polymorphic gene found by RFLP technique was cloned and sequenced.Lastly we analyzed the clinical data.Results To the CAPN 10 UC SNP43 polymorphism site of CAPN 10, 3 (1.18%) with the homozygote mutation, 47 (18.43%) with the heterozygote mutation were found in 255 typy 2 diabetes patients. The A allele frequency is 10.39% ,the G allele frequency is 89.31% in diabetes group. 1 (0.8%) with the homozygote mutation, 31 (24.35%) with the heterozygote mutation were foundin 125 healthy controls. The A allele frequency is 13.65% ,the G allele frequency is 86.35% in control group. There is no significiant differences between two groups in the A allele frequency (p>0.05). In diabetes group, HOMA-IR index is higher in CAPN10 UC SNP43 G/G genotype subjects than in CAPN10 UC SNP43 G/A and A/A genotype subjects. Compared between this two groups, it is seen the significiant differences in statistically. In diabetes group, we subsequently compared the clinical data between CAPN10 UC SNP43 G/G genotype subjects and CAPN10 UC SNP43 G/A and A/A genotype subjects,and found there is significiant differences in BMK P2PG^. TG between this two genotypes (p<0.05).Conclusions The data showed that there was UC SNP43 G/A polymorphism of calpain 10 gene in the Han nationality population in Tianjin of China, UC SNP43 G/A polymorphism of calpain 10 gene is associated with some indexes correlated insulin resistance. So it may partly cause insulin resistance of typy 2 diabetes. This polymorphism may be a genetic marker of typy 2 diabetes and insulin resistance in only parts of races or populations.
